From an endophytic Daldinia eschscholzii strain isolated from the agar-producing red alga Gracilaria sp. SGR-1, collected from the coast of South Sulawesi, Indonesia, a new lactone helicascolide C (1) was obtained as colourless crystals from the ethyl acetate extract together with the related structurally known compound helicascolide A (2). The structure of the new compound 1 reveals a carbonyl group replacing an alcohol group of compound 2.Thestructure of 1 was elucidated by X-ray diffraction and spectral analyses. Compound 1 showed fungistatic activity against the phytopathogenic fungus Cladosporium cucumerinum. (C) 2011 Phytochemical Society of Europe. Published by Elsevier B.V. All rights reserved.

Ibrahim Morgan, Ludger A. Wessjohann, Goran N. Kaluderovic

Summary: Anthraquinone derivatives are a class of compounds that exhibit various biological activities, including antifungal, antibacterial, antiviral, and anticancer activities. This study tested various synthetic anthraquinone derivatives against different cancer cell lines and found that most of them exhibit anticancer effects. One specific compound, 1,4-bis(benzyloxy)-2,3-bis(hydroxymethyl)anthracene-9,10-dione, exhibited the highest cytotoxicity against PC3 cells and was further investigated for its mechanism of action. The compound was found to induce apoptosis, disrupt cellular equilibrium, and cause cell cycle arrest.

Multicomponent Functionalization of the Octreotide Peptide Macrocyclic Scaffold

Aldrin V. Vasco, Leonardo G. Ceballos, Ludger A. Wessjohann, Daniel G. Rivera

Summary: The replacement of the disulfide bridge by other side chain linkages is a method for developing cyclic peptide drugs. This study describes a method on resin that enables the substitution of the disulfide bond with a tertiary lactam bridge and allows the addition of various exocyclic moieties. Conformational analysis in comparison with octreotide provides important information on the functionalization type for mimicking the bioactive peptide.

Purpurascenines A-C, Azepino-Indole Alkaloids from Cortinarius purpurascens: Isolation, Biosynthesis, and Activity Studies on the 5-HT2A Receptor

Yen T. H. Lam, Jana Hoppe, Quang N. Dang, Andrea Porzel, Alena Soboleva, Wolfgang Brandt, Robert Rennert, Hidayat Hussain, Mehdi D. Davari, Ludger Wessjohann, Norbert Arnold

Summary: Three new azepino-indole alkaloids, named purpurascenines A-C (1-3), along with a new-to-nature 7-hydroxytryptophan (4) and two known compounds, adenosine (5) and riboflavin (6), were isolated from the fruiting bodies of Cortinarius purpurascens. The structures of 1-3 were determined using spectroscopic analysis and ECD calculations. The biosynthesis of purpurascenine A (1) was investigated, and a biosynthetic route involving a direct Pictet-Spengler reaction between alpha-keto acids and 7-hydroxytryptophan (4) was proposed. Compound 1 showed no antiproliferative or cytotoxic effects against human cancer cells and exhibited some antagonistic effects on the 5-HT-dependent 5-HT2A activation and putative constitutive activity of the 5-HT2A receptor.
