Summary: Astragaloside VII (AST VII), a triterpenic saponin, has potential as a vaccine adjuvant due to its ability to promote a balanced Th1/Th2 immune response. However, the underlying mechanisms of its adjuvant activity have not been fully understood. This study investigated the effects of AST VII and its analogs on human whole blood cells and mouse bone marrow-derived dendritic cells (BMDCs). The results demonstrate that AST VII and its derivatives enhance pro-inflammatory responses, support dendritic cell maturation, and activate T cells in vitro.

Summary: A new natural product with pyrrolobenzodiazepine (PBD) framework, 7-methoxy-8-hydroxy cycloanthranilylproline (2), was isolated from marine-derived actinobacterium Streptomyces cacaoi 14CM034, along with cycloanthranilylproline (1). The compounds were structurally elucidated using various analytical techniques and showed notable antimicrobial activity. This study demonstrated the presence of PBD derivatives in S. cacaoi for the first time.

Summary: Salvia palaestina aqueous and methanol extracts were evaluated for in vitro anti-inflammatory properties, with methanol extract showing significant lipoxygenase inhibition. Ethyl acetate fraction from activity guided fractionation exhibited the best inhibition, and compounds responsible for the activity were identified as rosmarinic acid and methyl rosmarinate.

Summary: Polyether ionophores are a group of naturally occurring compounds mainly produced by Streptomyces species with various bioactivities. In this study, four undescribed and nine known polyether compounds were identified from Streptomyces cacaoi. The antimicrobial activity assays showed strong inhibitory effects of these compounds, including an undescribed compound, on the growth of Bacillus subtilis and methicillin-resistant Staphylococcus aureus (MRSA). Additionally, two compounds displayed significant cytotoxicity, with the C15-demethoxy derivative showing higher selectivity towards cancer cells than K41-A. Mechanistic experiments revealed that the uninvestigated compound acts as a multitarget compound, inhibiting autophagic flux, inducing reactive oxygen species formation, abolishing proteasome activity, and stimulating ER stress.

Summary: There is a high demand for novel approaches to treat age-related diseases, which are a major cause of death worldwide. Dysfunctional telomeres, which accelerate the aging process, are associated with many of these diseases. Telomerase, an enzyme responsible for maintaining healthy telomeres, has shown promise as a target for treatment. Natural products containing telomerase activators have protective and therapeutic effects against age-related diseases, particularly neurodegenerative, cardiovascular, and osteodegenerative disorders. However, the role of telomerase activation is often overlooked in studies.

Summary: Fifteen new neo-clerodane diterpenoids, including two undescribed glycosides, as well as a known iridoid glycoside and a phenylpropanoid glycoside, were isolated from the whole plant of Teucrium divaricatum subsp. divaricatum. Their structures were determined through spectral data analysis, and their anti-inflammatory and antimicrobial activities were evaluated. The compounds showed no significant antimicrobial activity against various bacterial and fungal strains, and were inactive up to the highest tested concentration of 50 μM on iNOS inhibitory activity.
