Personalised 3D Assessment of Trochanteric Soft Tissues Improves HIP Fracture Classification Accuracy

Alessandra Aldieri, Mara Terzini, Alberto L. Audenino, Cristina Bignardi, Margaret Paggiosi, Richard Eastell, Marco Viceconti, Pinaki Bhattacharya

Summary: The thickness of passive soft tissues surrounding the trochanteric region is related to hip fracture risk. Personalized measurement of thickness using computed tomography scans can improve the accuracy of fracture classification based on absolute risk values.

A theoretical analysis of the scale separation in a model to predict solid tumour growth

Barbara de Melo Quintela, Silvia Hervas-Raluy, Jose Manuel Garcia-Aznar, Dawn Walker, Kenneth Y. Wertheim, Marco Viceconti

Summary: This article discusses the factors affecting solid tumor growth and the importance of constructing personalized mathematical models. In order to address the wide range of scales in solid tumors, a multiscale model is proposed, which separates scales based on experimental and computational limitations.

Electron microscopy of cardiac 3D nanodynamics: form, function, future

Peter Kohl, Joachim Greiner, Eva A. Rog-Zielinska

Summary: This Review discusses the latest insight into the 3D nanostructure of the heart and its impact on cellular function during contraction and relaxation. Technological breakthroughs in biological sample preparation, 3D imaging, and data analysis have provided access to new data and driven the development of smart image-analysis algorithms. The authors highlight the scientific discovery and potential clinical relevance of understanding 3D cardiac nanodynamics, as well as predict further progress in the next decade.

Statistical Properties of a Virtual Cohort for In Silico Trials Generated with a Statistical Anatomy Atlas

Antonino A. La Mattina, Fabio Baruffaldi, Mark Taylor, Marco Viceconti

Summary: This study explored the use of statistical aliasing to expand patient cohorts while retaining original biomechanical characteristics. By simulating a clinical trial on a large cohort of virtual patients and monitoring biomarkers of interest, it offers a novel solution to address the limitations of prospective studies on osteoporosis-related hip fragility fractures.

A Credibility Assessment Plan for an In Silico Model that Predicts the Dose-Response Relationship of New Tuberculosis Treatments

Cristina Curreli, Valentina Di Salvatore, Giulia Russo, Francesco Pappalardo, Marco Viceconti

Summary: This study develops a computer simulation environment that can predict the dose-response of new therapeutic vaccines against tuberculosis, supporting the optimal design of clinical trials. Before using this in silico methodology, it is important to assess the credibility of the predictive model and a risk-informed credibility assessment plan is presented.

Effect of particularisation size on the accuracy and efficiency of a multiscale tumours' growth model

Vinicius Varella, Barbara de M. Quintela, Marek Kasztelnik, Marco Viceconti

Summary: This study explores the simplest particularisation/homogenisation scheme that can couple a model predicting the growth of a solid tumor with a tissue-scale model of cell-tissue biology. The study finds that even a simple algorithm can significantly reduce computational costs with negligible approximation errors.

Intra-operator Repeatability of Manual Segmentations of the Hip Muscles on Clinical Magnetic Resonance Images

Giorgio Davico, Francesca Bottin, Alberto Di Martino, Vanita Castafaro, Fabio Baruffaldi, Cesare Faldini, Marco Viceconti

Summary: This study evaluates the repeatability of manual segmentation of hip muscle volumes on clinical MRI data and finds that repeated manual segmentations are highly similar to each other. However, shape and surface similarity decrease when muscle extremities are included in the segmentations.

On the use of wearable sensors as mobility biomarkers in the marketing authorization of new drugs: A regulatory perspective

Marco Viceconti, Maria Tome, Wilhelmus Dartee, Igor Knezevic, Sabina Hernandez Penna, Claudia Mazza, Brian Caulfield, Judith Garcia-Aymerich, Clemens Becker, Walter Maetzler, Thierry Troosters, Basil Sharrack, Giorgio Davico, Solange Corriol-Rohou, Lynn Rochester

Summary: Loss of mobility is common in various health conditions and affects the quality of life. Recent advancements in technology now allow accurate monitoring of mobility in real-world settings, even for pathological populations. However, regulatory qualification is necessary before employing these methods in clinical trials. The Mobilise-D Consortium presents their experience with the European regulator, proposing a refined qualification strategy for using digital mobility outcome measures as biomarkers in drug trials.

Toward A Regulatory Pathway for the Use of in Silico Trials in the CE Marking of Medical Devices

Francesco Pappalardo, John Wilkinson, Francois Busquet, Antoine Bril, Mark Palmer, Barry Walker, Cristina Curreli, Giulia Russo, Thierry Marchal, Elena Toschi, Rossana Alessandrello, Vincenzo Costignola, Ingrid Klingmann, Martina Contin, Bernard Staumont, Matthias Woiczinski, Christian Kaddick, Valentina Di Salvatore, Alessandra Aldieri, Liesbet Geris, Marco Viceconti

Summary: In Silico Trials methodologies will have a significant impact on the development and risk reduction of medical devices in the future. The regulatory pathway for Digital Patient and Personal Health Forecasting solutions is clear, but more complex for In Silico Trials solutions. It is suggested that the European regulatory system should start an innovation process to avoid companies focusing on other markets like the USA.
