Summary: As oligonucleotides (ONs) and similar nucleic acid therapeutic modalities enter development pipelines, there is a constant need to develop bioanalytical methods to address their unique challenges. This report presents a toolbox of novel techniques that can be easily implemented for existing hybridization-ligation enzyme-linked immunosorbent assay formats, improving the sensitivity and specificity of these assays. These methods enhance the performance of bioanalytical assays, facilitating the research and development of innovative oligonucleotide therapeutics.

Summary: Recent studies have shown that enzymes undergo chemotaxis during catalysis. This study demonstrated the chemotaxis of zinc porphyrin as a cofactor mimic and its directed motions induced by ligand-protein binding. The dissociation constant for selected recognition was obtained by measuring the chemotactic shift, and a statistical thermodynamic model was derived to explain the directional migration of receptors.

Summary: The study utilized the CREST tool to calculate non-covalent ligand-receptor interactions and energy using the GFN2-xTB method, and blind predictions were made for the binding of 10 drug molecule ligands to the CB[8] receptor. Summary: Modulating the endocannabinoid system holds therapeutic potential in various pathophysiological processes. Novel Pyrrolo[2,1-c][1,4]benzodiazepines tricyclic compounds with high conformational similarity to the CB2 active site were designed for precise modulation. By synthesizing analogs and conducting binding assays, two potent and selective CB2 ligands were identified. Compound 4k exhibited oral bioavailability and favorable pharmacokinetic properties.

Summary: This study investigated the function and binding properties of two general odorant binding proteins (GOBPs) in Spodoptera frugiperda. It was found that SfruGOBP1 had binding affinities to a few volatiles and insecticides, while SfruGOBP2 showed a broader ligand-binding spectrum, suggesting its importance in perceiving host volatiles. Molecular docking and site-directed mutagenesis assays provided insight into the key amino acid residues involved in the binding of insecticides to the GOBPs. This study offers valuable information for understanding the olfactory mechanism of GOBPs in S. frugiperda.

Summary: Cytochrome c oxidase (CcO) is a hemeprotein that reduces dioxygen to water. It has a unique structure with a binuclear center composed of a copper atom and a heme iron. In the CO complex of CcO, the CO molecule dissociates from the heme iron and moves to a temporary binding site between the copper atom and the heme iron. This new structure provides insight into the ligand dissociation trajectory and protein dynamics.

Summary: Hyphantria cunea is an invasive pest species in China that uses type II sex pheromone components. In this study, the binding mechanisms of its sex pheromone components to their respective pheromone-binding proteins were explored. The results showed that HcunPBPs have different selectivities for pheromone components with major and minor structural differences, providing insight into the olfactory mechanism of moths using type II pheromone components.

Summary: In this study, the odorant binding protein (OBP) gene family in Dioryctria abietella was investigated. Expression profiles showed that most OBPs were highly expressed in the antennae of females. Two DabiOBPs were isolated from the antennae and exhibited different odorant response spectra. Molecular simulations revealed key residues involved in the interactions of DabiOBPs and ligands. This study highlights the olfactory roles of antennal DabiOBPs in D. abietella and helps in the identification of potential behavioral compounds for pest control.
