Summary: Research has shown that porous particles of appropriate size and porosity can accumulate in hair follicles and furrows on the skin, providing a potential area for drug delivery. The combination of follicle orifices and furrows can cover up to 20% of the skin surface, making them valuable for optimizing bioavailability of topical formulations. This work highlights the potential of skin furrows for topical drug delivery.

Summary: This study evaluated the effectiveness of omiganan in the treatment of facial seborrheic dermatitis. The results showed that omiganan did not significantly improve clinical symptoms or other biomarkers. In contrast, ketoconazole significantly reduced disease severity and had positive effects on Malassezia abundances, microbial diversity, and skin barrier function. The importance of evaluating omiganan in the treatment of facial seborrheic dermatitis is limited.

Summary: This study finds that cabazitaxel (CTX) can be dissolved in alpha-tocopherol at high concentrations exceeding 100 mg mL(-1). The addition of 30% dimethyl sulfoxide (DMSO) to the alpha-tocopherol/CTX solution improves its permeation through human skin. Topical application of CTX using a DMSO and alpha-tocopherol solvent shows significant suppression of tumor growth in nude mice bearing skin cancer xenografts.

Summary: The evaluation of bioequivalence (BE) for topical dermatological drug products is challenging, and the aim of this study was to evaluate the use of Confocal Raman Spectroscopy (CRS) for BE assessment. Two commercially available formulations were found to deliver similar amounts of drug across the skin in vitro and resulted in comparable skin uptake values in vivo. This is the first study to demonstrate the capability of CRS for the evaluation of BE for topical products.

Summary: A neutral oil-incorporated liposomal system (LOS) was developed to enhance the skin absorption of ascorbic acid (Vit C). Sodium deoxycholate as an edge activator and camellia oil as a neutral oil were found to significantly improve the absorption of Vit C. This optimized lipid nanocarrier shows promise in promoting the skin absorption and dermatological functions of Vit C.

Summary: In this study, novel fatty acid vesicles loaded with magnolol were prepared using 10-hydroxy decanoic acid (HDA) and oleic acid (OA) for skin cancer treatment. The performance of different formulations was statistically evaluated in terms of particle size, polydispersity index, zeta potential, and entrapment efficiency. Both in vitro and in vivo experiments demonstrated that HDA-based vesicles exhibited better drug permeation and skin deposition, and showed superior efficacy in attenuating DMBA-induced skin cancer.

Summary: The main goal of medicine is to achieve the best possible outcome for therapy. Nanocarriers and co-delivery strategies have been invented to meet this need, benefiting not only cancer treatment but also many other clinical issues, especially skin diseases. This review aims to summarize the research state of nanocarriers and co-delivery strategies for topical agents' applications targeting skin diseases. The challenge for future medicine lies in delivering drugs with spatial and temporal control, and the co-encapsulation of drugs and appropriate administration forms are important and unmet needs.
