Summary: In this study, a novel KDF1 variant associated with human tooth agenesis was identified. Functional analysis showed that KDF1 variants inhibited the activation of canonical Wnt signaling. Furthermore, phenotypic analyses revealed tooth and gingival epithelial developmental anomalies related to the KDF1 variant. These findings broaden the phenotypic spectrum of KDF1-related disorders and provide new evidence for the crucial role of KDF1 in regulating human dental and oral epithelial development.

Summary: This study aimed to evaluate associations between severe early childhood caries (S-ECC), the oral microbiome, and variants in genes involved in taste sensation. Results showed significant associations between three loci in the PLCB2 gene and S-ECC, as well as associations between variants in other genes and dental biofilm composition. These findings contribute to our understanding of the role of genetics in S-ECC.
