Beneficial effect of dehydroepiandrosterone on pulmonary hypertension in a rodent model of pulmonary hypertension in infants

BACKGROUND: Pulmonary hypertension (PH) is a disease that affects the adult or infant population. Dehydroepiandrosterone (DHEA), a steroid hormone, has been previously shown to prevent and to reverse PH in an adult rat model. We thus investigated its effect in a rat-pup model of chronic hypoxic PH. METHODS: Animals were maintained for 3 wk in a hypobaric chamber to induce PH, with or without concomitant treatment with DHEA (30 mg/kg every alternate day). RESULTS: DHEA significantly reduced mean pulmonary artery pressure (measured by right cardiac catheterization), pulmonary artery remodeling (evaluated by histology), and right-ventricular hypertrophy (measured by echography and by the Fulton index). At the level of the pulmonary artery smooth muscle cell (PASMC), DHEA increased activity and expression of the large-conductance Ca2+-activated potassium channel (BKCa) (assessed by means of the patch clamp technique). DHEA also inhibited both serotonin- and KCI-induced contraction and smooth muscle cell proliferation. CONCLUSION: Collectively, these results indicate that DHEA prevents PH in infant rats and may therefore be clinically relevant for the management of PH in human infants.

Summary: Stilbene metabolites are of great interest due to their similar or even stronger biological effects compared to their parent compounds. Their study is highly relevant as they are predominant in biological fluids. The metabolism of oxyresveratrol (ORV) and gnetol (GN) resulted in the formation of several glucuronide metabolites, which showed the ability to decrease the production of inflammatory factors in stimulated macrophages.

Involvement of the Piezo1 and TRPV4 stretch-activated channels in pulmonary hypertension

Summary: Pulmonary hypertension is a pulmonary circulation pathology characterized by remodeling and hyperreactivity of the pulmonary arteries. The vasodilatation/vasoconstriction balance is altered, favoring constriction due to the proliferation of smooth muscle cells and the development of endothelial dysfunction. In addition, mechanical forces cause modifications in the pulmonary arteries, leading to changes in the activation of stretch-activated channels such as Piezo1 and TRPV4. These channels, sensitive to stretch, can induce various cellular physiological responses, which significantly contribute to the development and continuation of the pathology.

Summary: This study compared the toxic effects of nickel oxide nanoparticles (NiONPs) on human pulmonary artery endothelial cells (HPAEC) under physiological and pathological conditions. The results showed that under pathological conditions, NiONPs had a more pronounced adverse effect on oxidative stress, inflammation, calcium signaling, and mitochondrial function in the cells compared to physiological conditions.

Scorpion venom is composed of enzymes and peptides, and different species have different medical importance. France does not have scorpion antivenom as scorpion stings are generally not fatal.

Genetically-encoded BRET probes shed light on ligand bias-induced variable ion selectivity in TRPV1 and P2X5/7

Summary: This study developed highly sensitive bioluminescence resonance energy transfer (BRET) probes to track selective ion permeability in ion channels. The results showed that well-known TRPV1 inhibitors more potently inhibit Ca2+ influx than K+ efflux induced by capsaicin (CAPS). Furthermore, AMG517 was found to be a partial agonist of K+ efflux and triggered TRPV1-dependent cell membrane hyperpolarization. Ligand biases were also observed in other families of cationic channels.

Pathophysiological Roles of the TRPV4 Channel in the Heart

Summary: The TRPV4 channel plays a role in intracellular calcium handling in cardiac cells and is involved in various physiological functions in the cardiovascular system. It modulates mitochondrial activity, calcium homeostasis, electrical activity and contractility of cardiomyocytes, as well as vascular permeability, dilatation and constriction. Furthermore, TRPV4 channels are implicated in cardiac pathological processes such as fibrosis, hypertrophy, ischemia-reperfusion injuries, heart failure, myocardial infarction and arrhythmia. This manuscript provides an overview of the implications of TRPV4 channel in cardiac physiology and discusses its potential as a therapeutic target for cardiovascular diseases.
