Background Prenatal maternal smoking and prematurity independently affect wheezing and asthma in childhood. Objective We sought to evaluate the interactive effects of maternal smoking and prematurity upon the development of early childhood wheezing. Methods We evaluated 1,448 children with smoke exposure data from a prospective urban birth cohort in Boston. Maternal antenatal and postnatal exposure was determined from standardized questionnaires. Summary: This study identified a functional variant at the 17q12-21.1 locus associated with early-onset asthma in African American individuals. The lead signal, rs11078928, was found to be the top association for early-onset asthma after meta-analysis across cohorts. Additionally, a haplotype analysis suggested that the asthma association most closely with this variant.

Adherence rates during a randomized controlled trial evaluating the use of blinded acetaminophen and ibuprofen in children with asthma

Summary: The study showed that over 85% of doses of acetaminophen or ibuprofen were adherence to the protocol of blinded study medications, while less than 15% were deviations to OTC medications. Better adherence was observed in the ibuprofen group compared to the acetaminophen group. In individual cases, over 75% of participants remained fully adherent by not receiving any OTC medications.

Summary: Pharmacogenetic studies in asthma cohorts have identified loci associated with response to medications in individuals of European descent, but differences exist in individuals of African descent. This study aimed to understand pharmacogenetic mechanisms regulating therapeutic responsiveness in people of African descent and detected novel significant loci.

Summary: The allergy section of the electronic health record plays a crucial role in clinical decision-making and treatment planning, but current allergy modules do not meet the requirements. We recommend collaboration between EHR vendors and allergists to optimize and modernize allergy documentation, providing meaningful, accurate, timely, and comprehensive allergy information.
