Leishmanicidal activity of the Agaricus blazei Murill in different Leishmania species

Leishmaniasis is a major public health problem, and the alarming spread of parasite resistance underlines the importance of discovering new therapeutic products. The present study aims to investigate the in vitro leishmanicidal activity of an Aga ricus blazei Murill mushroom extract as compared to different Leishmania species and stages. The water extract proved to be effective against promastigote and amastigote-like stages of Leishmania amazonensis, L chagasi, and L major, with IC50(50% inhibitory concentration) values of 67.5, 65.8, and 56.81.1g/mL for promastigotes, and 115.4, 112.3, and 108.4 pg/mL for amastigotes-like respectively. The infectivity of the three Leishmania species before and after treatment with the water extract was analyzed, and it could be observed that 82%, 57%, and 73% of the macrophages were infected with L amazonensis, L major, and L chagasi, respectively. However, when parasites were pre-incubated with the water extract, and later used to infect macrophages, they were able to infect only 12.7%, 24.5%, and 19.7% of the phagocytic cells for L amazonensis, L chagasi, and L major, respectively. In other experiments, macrophages were infected with L amazonensis, L chagasi, or L major, and later treated with the aforementioned extract, presented reductions of 84.4%, 79.6%, and 85.3% in the parasite burden after treatment. A confocal microscopy revealed the loss of the viability of the parasites within the infected macrophages after treatment with the water extract. The applied extract presented a low cytotoxicity in murine macrophages and a null hemolytic activity in type O+ human red blood cells. No nitric oxide (NO) production, nor inducible nitric oxide syntase expression, could be observed in macrophages after stimulation with the water extract, suggesting that biological activity may be due to direct mechanisms other than macrophage activation by means of NO production. In conclusion, the results demonstrate that the A blazei Murill water extract can potentially be used as a therapeutic alternative on its own, or in association with other drugs, to treat Visceral and Cutaneous Leishmaniasis. Summary: Visceral leishmaniasis (VL) is a fatal disease caused by Leishmania infantum, and dogs are the main reservoirs. A vaccine against canine visceral leishmaniasis (CVL) is important in controlling human and CVL infections by reducing L. infantum infection. This study evaluated the potential of two polymeric nanoformulations loaded with Leishmania amazonensis antigens as a VL vaccine using golden hamsters as a model. The results showed that the vaccines had no adverse reactions and significantly reduced the parasitic load in the liver and spleen. These vaccines may be promising candidates for a VL vaccine.

Summary: Evidence suggests that there may be racial differences in risk factors associated with the development of Alzheimer's disease and related dementia (ADRD). Whole-genome sequencing analysis revealed a novel combination of three pathogenic variants in a Peruvian family with a strong clinical history of ADRD. In silico and in vitro studies indicated that these variants may affect the neurovascular unit and key molecular pathways associated with dementia spectrum disorders.

Summary: Background: Visceral leishmaniasis (VL) is a critical public health problem in over ninety countries, and control measures in Brazil have been insufficient. A chimeric protein called rMELEISH was developed for the accurate diagnosis of VL in dogs and humans. In trials, rMELEISH showed 100% sensitivity and specificity in distinguishing VL cases from other diseases and healthy samples. These preliminary results suggest that rMELEISH has potential for future diagnostic studies in the field.

Summary: This study aimed to develop a new recombinant multiepitope protein, rTC, for the diagnosis of Chagas disease. The results showed that rTC could differentiate between indeterminate and cardiac cases, as well as cross-reactive diseases and healthy samples, with 98.28% sensitivity and 96.67% specificity, respectively. These findings suggest the potential of rTC to be tested in future studies against a larger serological panel for Chagas disease diagnosis.

A Systematic Review and Meta-Analysis Comparing the Diagnostic Accuracy Tests of COVID-19

The study highlights the need to improve the performance of these diagnostic tests to effectively control COVID-19 outbreaks.

Summary: Our study used an immunotherapeutic approach combining a vaccine and drug to successfully treat mice infected with Leishmania parasites. This approach significantly reduced the parasite load in the animals' internal organs and increased the production of protective antibodies. The inclusion of the vaccine and adjuvant also reduced the toxicity of the drug.

Summary: The treatment of visceral leishmaniasis (VL) is limited by issues such as drug toxicity, high cost, and resistant strains. This study evaluated an immunotherapy protocol using a combination of recombinant LiHyp1, monophosphoryl-lipid A (MPLA), and amphotericin B (AmpB) in a murine model. Results showed that this combination led to the lowest parasite burden, higher levels of protective immune response, and minimal organ damage. These findings suggest that the rLiHyp1/MPLA/AmpB combination could be a potential immunotherapy protocol for VL treatment.

In this study, a novel recombinant chimeric antigen (N4S11-SC2) was developed using specific B-cell epitopes from the SARS-CoV-2 virus. The N4S11-SC2 antigen showed high sensitivity and specificity in detecting specific antibodies in serum and urine samples, making it a potential alternative marker for diagnosing SARS-CoV-2 infection.

Summary: The dynamics of anti-T. gondii antibodies and seroconversion in naturally infected goats from the last third of pregnancy to 100 days of lactation were evaluated, and their relationship with hematological and dehydration parameters was examined. Current drugs on the market are often toxic with numerous side effects, necessitating the search for new, more potent molecules. Medicinal plants have been found to contain a wide range of novel molecules that can effectively combat different types of helminths. This review provides an overview of the methods, activities, and mechanisms of action of cyclic peptides, specifically cyclotides, for treating helminthic infections.
