Objectives: Time trends in the incidence of pancreatic cancer vary considerably between countries. The aim of this study was to provide time trends in incidence during a 25-year period in a well-defined French population. Methods: The cancer registry in Burgundy (France) was used to study time trends between 1981 and 2005 by sex, age, subsite, and histology. They were analyzed using an age-period cohort model. Results: Age-standardized incidence rates increased from 5.7 (1981-1985) to 7.9 per 100,000 (2001-2005) in men and from 2.6 to 4.6 in women. The mean percentage of variation by the 5-year period was +9.9% (P < 0.001, 95% confidence interval, 6.2%-13.6%) and +13.4% (P = 0.004, 95% confidence interval, 9.4%-17.5%), respectively. The increase in incidence was higher for cancers of the tail and corpus than for cancers of the head of pancreas and for malignant pancreatic endocrine tumors than for adenocarcinomas. The cumulative risk of developing a pancreatic cancer rose from 0.51% for men born in 1900 to 1.13% for those born in 1950. It was 0.34% and 0.55% for women, respectively. Conclusions: The incidence of pancreatic cancer has increased sharply in France both by period and by birth cohort. Etiological studies are required to explain the increase in the incidence.

Summary: This study analyzed the potential effects of colorectal cancer screening programs in several European countries. The results showed that the proportion of screen-detected colorectal cancers varied widely between countries. Screen-detected cancers were more likely to be detected at an early stage compared to non-screen-detected cancers.

Summary: The study examines global survival trends in leukemia among young patients aged 0-24 years, revealing significant variations in survival rates across different countries and regions. Children consistently have higher survival rates compared to adolescents and young adults.

Health status of prevalent cancer cases as measured by mortality dynamics (cancer vs. noncancer): Application to five major cancers sites

Summary: This study proposes a statistical approach to describe the health status of patients with different types of cancer in France by estimating the probabilities of death among prevalent cases. The study finds that most deaths from cancer occur within the first 5 years after diagnosis, but the probability of death from cancer decreases with increasing time since diagnosis, except for breast cancer. The time at which the probability of death from cancer becomes lower than that from other causes varies depending on age and cancer site.

Clear Improvement in Real-World Chronic Myeloid Leukemia Survival: A Comparison With Randomized Controlled Trials

Claudia Vener, Silvia Rossi, Pamela Minicozzi, Rafael Marcos-Gragera, Helene A. Poirel, Marc Maynadie, Xavier Troussard, Gabriella Pravettoni, Roberta De Angelis, Milena Sant

Summary: Tyrosine kinase inhibitors (TKIs) have improved the prognosis of patients with chronic myeloid leukemia (CML). Comparing survival data from population-based studies and randomized control trials (RCTs), it was found that the prognostic improvement highlighted by RCTs was confirmed in real-world settings. However, some European countries still have lower survival rates, possibly due to delayed use of TKIs.

Summary: This study evaluates the prognostic performance of CDX2 and CD3 biomarkers in colon cancer patients, finding that they have prognostic value in stage III but not in stage II. The combined effect of CDX2 and CD3 helps identify a subgroup of stage III patients with a particularly good prognosis, and this association is feasible in routine pathology laboratories.

Summary: This study provides a comprehensive examination of worldwide variation and trends in survival from brain tumors in adults by analyzing individual data from 2000 to 2014. The results show significant regional differences and temporal changes in survival rates, especially for the most common sub-types, with remarkable improvements since 2005 due to the uptake of chemoradiation. However, there are still some countries that lag behind in terms of survival rates.

Integrative Clinical and DNA Methylation Analyses in a Population-Based Cohort Identifies CDH17 and LRP2 as Risk Recurrence Factors in Stage II Colon Cancer

Benjamin Tournier, Romain Aucagne, Caroline Truntzer, Cyril Fournier, Francois Ghiringhelli, Caroline Chapusot, Laurent Martin, Anne Marie Bouvier, Sylvain Manfredi, Valerie Jooste, Mary B. Callanan, Come Lepage

Summary: Using DNA methylation-based screening, researchers have developed a classifier that can segregate stage II colon cancer into four sub-classes. They identified hypermethylation at specific CpG sites in CDH17 and LRP2 genes that independently confer increased or reduced risk of cancer recurrence. Functional enrichment and immune cell infiltration analyses revealed distinct molecular features in each sub-class.

Survival variability across hospitals after resection for pancreatic adenocarcinoma: A multilevel survival analysis on a high-resolution population-based study

Summary: The surgical volume of hospitals has a significant impact on the survival rate of patients with pancreatic ductal adenocarcinoma (PDAC). High-volume hospitals have better survival rates, while low-volume hospitals have higher post-operative mortality. The centralization of pancreatic surgery is a complex issue.

Biliary tract cancers have distinct epidemiological patterns and clinical characteristics according to tumour site

Francois Ghiringhelli, Valerie Jooste, Sylvain Manfredi, Audrey Hennequin, Come Lepage, Anne -Marie Bouvier

Summary: This study investigated the epidemiology, treatment, and prognosis of biliary tract cancers over the past decade. The incidence of biliary tract cancers did not vary significantly, except for a slight increase in intrahepatic cholangiocarcinoma in men and a decrease in ampulla cancer. Age, performance status, and hospital type were associated with the likelihood of surgery. Ampulla cancer had higher survival rates compared to other sites.

Estimating complete cancer prevalence in Europe: validity of alternative vs standard completeness indexes

Elena Demuru, Silvia Rossi, Leonardo Ventura, Luigino Dal Maso, Stefano Guzzinati, Alexander Katalinic, Sebastien Lamy, Valerie Jooste, Corrado Di Benedetto, Roberta De Angelis

Summary: Comparable indicators on complete cancer prevalence are needed in Europe. Two alternative approaches, empirical indices derived from European registries and pre-calculated indices derived from US-SEER cancer registries, were compared to standard estimates using data from 62 registries in 27 countries. Empirical estimates differ little from standard estimates for elderly-diagnosed tumors, but may underestimate complete prevalence for early-onset cancers. SEER estimates are comparable to standard ones for most cancers, even derived from short time series, but longer observations are needed for certain cancers with remarkable differences in incidence and prognosis between European and US populations.
