Induction of pro-inflammatory gene expression by Escherichia coli and mycotoxin zearalenone contamination and protection by a Lactobacillus mixture in porcine IPEC-1 cells

This work investigated the effect of Escherichia coli K88 and zearalenone contamination on pro-inflammatory gene expression (Toll like receptors, cytokines) and signalling molecules and the protective activity of a mixture of Lactobacilli sp. (Lactobacillus plantarum, Lactobacillus acidofilus and Lactobacillus paracasei) in porcine intestinal epithelial cells as part of the local immune system. IPEC-1 cell monolayer was exposed for 1 h to the individual or combined action of E. coli, zearalenone and lactobacilli mixture. Our results showed that TLRs (1-10) and cytokine (IL-1,-6,-8,-10, TNF-alpha, IFN-gamma) genes expressed early (after 1 h of culture) in IPEC-1 cells. E. coli alone increased the TLRs mRNA expression, especially TLR4 and the inflammatory cytokines while ZEA alone showed either no effect or a marginally effect on TLRs, cytokines, and signalling genes when compared to untreated cells. The combined actions of the two contaminants lead to a synergistically up-regulation of key cytokines (IFN-gamma, IL-10 and TNF-alpha) and TLRs (-2,-3,-4,-6, and -10). The live lactobacilli mixture was able to attenuate the pathogen and mycotoxin-induced response by downregulated the majority of inflammatory related genes suggesting that this mixture has an immunomodulatory potential and may be used to lower the inflammatory response. Assessment of the ability of dietary yeast-fermented rapeseed meal to modulate inflammatory and oxidative stress in piglets after weaning

Summary: The aim of this study was to investigate the potential of a diet containing rapeseed meal fermented by Saccharomyces cerevisiae to reduce intestinal inflammation, diarrhoea and oxidative stress in piglets after weaning. The results showed that the fermented rapeseed meal diet had a positive effect on piglet intestinal health, reducing the incidence of diarrhoea and improving antioxidant levels.

Co-Contamination of Food and Feed with Mycotoxin and Bacteria and Possible Implications for Health

Summary: Food and feed safety is a major concern for both animal and human health, as contamination with pathogens can occur frequently. This study investigated the effects of simultaneous contamination with mycotoxins and bacterial toxins on cell viability, oxidative stress, and inflammation. The results showed significant additive/synergistic effects of co-exposure to fungal and bacterial toxins, suggesting that the concomitant contamination with bacteria and mycotoxins could lead to higher toxicity.

Summary: The study evaluated the response of grape seed meal byproduct (GSM) to oxidative stress induced by E. coli lipopolysaccharide (LPS) and dextran sulphate sodium (DSS) in vitro and in vivo. The results showed that GSM possessed antioxidant properties and could counteract the oxidative stress response induced by LPS and DSS, as well as restore the levels of endogenous antioxidant enzymes in the colon and lymph nodes. These beneficial effects were mediated through the Nrf2 signaling pathway.

Summary: This study investigates the proteomic effects of mycotoxins on piglets' liver and the potential compensatory effects of grape seed and sea buckthorn meal as dietary antioxidants. It found that mycotoxins affect metabolism, antioxidant capacity, and various metabolic pathways in piglets' liver, and antioxidants partially restore the expression of affected proteins. This research is of great significance for protecting the health of farm animals and the quality of meat consumed by humans.

Using In Silico Approach for Metabolomic and Toxicity Prediction of Alternariol

Summary: In this study, an in silico working model based on online computational programs was used to predict the Phase I and II metabolites of alternariol and provide a detailed ADME/Tox profile. A total of 12 metabolites were identified corresponding to the metabolomic profile of alternariol. The ADME profile indicated moderate or high intestinal absorption probability but low probability to penetrate the blood-brain barrier. The computational model also predicted other toxicological endpoints for the compounds, such as vascular toxicity, haemato-toxicity, diarrhea, and nephrotoxicity.

Effects and Underlying Mechanisms of Zearalenone Mycotoxin at Concentrations Close to the EC Recommendation on the Colon of Piglets after Weaning

Summary: Zearalenone (ZEN), a mycotoxin produced by Fusarium fungi, contaminates food and feed, causing health issues for humans and animals. Pigs, as major consumers of grains, are particularly sensitive to mycotoxins. This study observed the effects of ZEN at concentrations below and above the EU recommendation on oxidative and inflammatory responses in the colon of weaned piglets. Exposure to the lower concentration of ZEN (75 ppb) did not result in changes in stress and inflammation markers or associated signaling pathways.
