Summary: This retrospective study compared bone mineral density changes in postmenopausal women with severe osteoporosis after stopping teriparatide therapy and receiving denosumab or bisphosphonates treatment for 12 months. The results showed that after 12 months, sequential denosumab treatment appeared to yield higher additional lumbar spine bone mineral density gain compared with bisphosphonates treatment.

Summary: Osteoporotic fractures have significant impact on quality of life, but half of the patients may discontinue their anti-osteoporosis medication. Factors associated with medication discontinuation include younger age, female gender, non-use of calcium supplements, and anti-resorptive medication.

Summary: This study investigated circulating miRNAs as potential biomarkers for monitoring treatment response in women with postmenopausal osteoporosis on denosumab therapy. The results showed significant changes in expression of seven miRNAs after two years of treatment, which correlated with changes in bone mineral density and bone turnover markers.

Summary: The study developed a clinical decision support system at the University of Ferrara, Italy, incorporating a machine learning system for providing suggestions on osteoporosis prevention and treatment to physicians. The experiment results showed satisfactory reliability of the system, with accuracies reaching around 90% in some cases.

Summary: Osteoporosis is a systemic bone disease characterized by reduced bone mass and deterioration of bone microarchitecture. Conventional anti-osteoporotic pharmaceutics have side effects, leading many women to seek botanicals as an alternative therapy. Plant-derived compounds are being increasingly researched as potential maintenance for bone health in perimenopausal and postmenopausal women.

Summary: MicroRNAs play a regulatory role in bone homeostasis, and circulating microRNAs are considered as potential biomarkers for bone health. This study investigated the impact of anti-osteoporotic treatments on circulating microRNAs in OVX and SHAM rats, revealing that these treatments can reverse bone loss and modulate specific miRNA expressions. The findings suggest that bone-related miRNAs could serve as promising biomarkers for monitoring treatment efficacy in osteoporosis.

Summary: Osteoporosis often coexists with metabolic diseases in postmenopausal women, suggesting shared pathogenetic mechanisms. Anti-osteoporotic medications may affect the pathogenesis of some metabolic diseases, while medications for metabolic diseases may adversely affect bone metabolism. Understanding the links between osteoporosis and metabolic diseases may have clinical implications.

Summary: Objective of this study was to identify significant ferroptosis-associated biomarkers for postmenopausal osteoporosis (PMOP) and establish a diagnostic and personalized treatment approach. The results showed that a ferroptosis-associated gene signature can be used for early diagnosis of PMOP and two different subtypes of PMOP were identified.

Summary: The study found that a combination treatment of teriparatide and denosumab in postmenopausal osteoporosis patients leads to greater increase in bone density compared to monotherapy, without significant difference in adverse events.

Summary: This study investigated risk factors for denosumab discontinuation in patients with postmenopausal osteoporosis (PMO). The results showed that a low BMI, low serum Alb levels, and a high number of vertebral fractures were independent risk factors for denosumab discontinuation.

Summary: The purpose of this study was to retrospectively examine predictors of fracture risk when adult patients experienced a denosumab treatment lapse or discontinuation in a real-world clinic setting. Off-treatment status may be associated with a higher risk of fractures. Dental problems and logistical barriers were the most common reasons for lapses and discontinuations.

Summary: The study found that the majority of patients on DMTs switched to alternative medications within 6 months. Reasons for stopping the drugs were mainly lack of efficacy, lack of tolerance, and side effects. The proportion of patients on most DMTs remained stable until 2014 and 2015, with a slight decrease noted thereafter.

Summary: The duration of denosumab treatment significantly influences the efficacy of subsequent zoledronate infusion in maintaining bone mineral density gains. Frequent follow-up of patients treated with denosumab for more than 3 years is advisable as additional therapeutic interventions may be necessary.

Summary: Bone targeted delivery of estrogen using citric acid modified diblock copolymer poly(2-ethyl-2-oxazoline)-poly(e-caprolactone) (CA-PEOz) drug delivery system significantly increased bone targeting and achieved rapid drug release under bone acidic conditions. This system reduced bone loss in postmenopausal osteoporotic mice and minimized estrogenic side effects on the uterus. The study demonstrates the potential of citric acid as an effective bone targeting modifier molecule and provides a new approach for bone-targeted delivery of estrogen for the treatment of postmenopausal osteoporosis.
