Summary: This study identifies SIK2/SIK3 as potential drug targets for treating osteoporosis and successfully develops an orally available SIK2/SIK3 inhibitor. The inhibitor stimulates bone formation and increases bone density without apparent toxicity. These findings provide a new approach for the pharmacological treatment of osteoporosis.

Summary: This meta-analysis evaluated the evidence for the use of parathyroid hormone (PTH) analogues to improve fracture healing. PTH improved functional outcomes but did not affect fracture healing rate or reduce pain. The evidence for PTH analogue use in fracture healing is less clear in younger, non-osteoporotic patient populations, warranting further research.

Summary: Low vitamin D levels are common in patients with hip fractures. Patients with low vitamin D have higher risks of delirium, new hip fractures, and medical readmissions, but not orthopedic complications. Further studies are needed to investigate the role of vitamin D supplementation in preventing orthopedic complications.

Summary: This study examined the long-term survival and incidence of secondary fractures after fragility hip fractures. The 5-year survival rate was 62%, with higher mortality risk in patients with GNRI < 92. The 5-year incidence of secondary fractures was 22%, which was significantly higher in patients with a BMI < 20.

Summary: Data on vitamin D status of patients with inherited ichthyosis in Europe is limited, with deficiencies found to be more pronounced in specific genetic subtypes. Low 25(OH)D3 levels were associated with severity of scaling, highlighting the importance of screening and substituting for vitamin D deficiency in ichthyosis patients according to recent guidelines.

Summary: The prevalence of secondary hyperparathyroidism (SHPT) is high in patients undergoing bariatric surgery, with a significant number of patients experiencing persistent SHPT for more than 1 year. The current vitamin D3 and calcium supplementation protocols may not effectively prevent SHPT in post-bariatric patients. Further studies are needed to develop effective treatment strategies to mitigate the adverse effects of bariatric surgery on bone metabolism.

Summary: This study investigated the relationship between 25-hydroxyvitamin D and 1,25-dihydroxyvitamin D with intact parathyroid hormone in outpatient population in Germany. The results showed an inverse correlation between 25-hydroxyvitamin D and intact parathyroid hormone, which was independent of age, sex, creatinine, calcium, and phosphate. However, 1,25-dihydroxyvitamin D was only positively associated with intact parathyroid hormone in women and in the subgroup with lower 25-hydroxyvitamin D.

Summary: The study showed that PTH and Scl-Ab can decrease the expression of miR-19a/b in bones, while antagonizing miR-19a/b can increase bone mass. Furthermore, antagonizing miR-19a/b enhances bone mass gain and restores bone loss by supporting bone formation and decreasing bone resorption.

Summary: This study analyzed the association between the prescription of active vitamin D and fracture incidence in hemodialysis patients. The results suggested that active vitamin D does not have a PTH-independent benefit in fracture prevention, supporting the current guideline of using active vitamin D only in patients with elevated or rising PTH.

Summary: Axial spondyloarthritis is a chronic inflammatory rheumatic disease that affects the axial skeleton and causes severe pain and disability. Early diagnosis and appropriate treatment can reduce the severity of the disease and the risk of progression. Different types of drugs, such as bDMARDs and JAK inhibitors, are available for treatment. However, treatment failure is common and requires individualized treatment strategies.

Summary: The TEAM Project analyzed the transition of patients with metabolic bone diseases from pediatric to adult care in Spain and developed consensus recommendations. A total of 86 physicians participated, reaching consensus on 45 statements. There were disagreements regarding the appropriate and feasible transition time and model. Lack of resources and coordination between pediatric and adult units were identified as the main barriers for successful transition.
