Identification of small molecular compounds and fabrication of its aqueous solution by laser-ablation, expanding primordial cartilage

Objective: The discovery of small molecular compounds that expand cartilage is needed. We searched for small molecular compounds that expand cartilage or enhance the actions of bone morphogenetic proteins (BMPs) on cartilage. Design: Metatarsal primordial cartilage explants prepared from 14.5 days postcoitum (d.p.c.) mouse embryos were organ-cultured in the presence or absence of BMPs and/or 4-(5-Benzol[1,3]dioxol-5-yl-4-pyrldin-2-yl-1H-imidazol-2-yl)-benzamide hydrate (BPIB) and its related molecules. The perichondrium was removed from some of the cartilage explants by partial digestion with collagenase. BPIB aqueous solution was prepared by fragmenting BPIB crystals in water with laser irradiation and then added to cartilage explants in organ culture. Results: We found that small molecular compounds, BPIB, available as SB431542 from Sigma and its related molecules, expand primordial cartilage explants in organ culture. These molecules are transforming growth factor-beta (TGF-beta) inhibitors, and the addition of excess TGF-beta reduced cartilage expansion induced by these molecules. The co-administration of BPIB and BMPs synergistically expanded cartilage explants. Removal of the perichondrium abolished BIPB-induced cartilage expansion but not BMP-induced cartilage-expansion, suggesting that BPIB, but not BMPs, expands cartilage through the perichondrium. Furthermore, we used the laser-ablation technique to generate BPIB aqueous solution in the presence of 2-hydroxypropyl-beta-cyclodextrin (HP-beta-CD) without the use of hazardous dimethyl sulfoxide (DMSO). The laser-ablation-generated BPIB aqueous solution was more stable, expanded cartilage explants more effectively than BPIB colloidal solution prepared with DMSO, and synergistically enhanced BMP-induced cartilage expansion. Conclusions: A small molecular compound, BPIB, expands primordial cartilage explants. A BPIB aqueous solution was created by laser-ablation without using DMSO and proved to be biologically active. Growth Enhancement of Organic Nonlinear Optical Crystals by Femtosecond Laser Ablation

Ablation can generate new crystal steps on the surface, but does not deteriorate the nonlinear optical properties. Controlling crystal shape through damage-less ablation process may potentially enhance the nonlinear optical properties of organic crystals.

L-serine polymorphism controlled by optical trapping with high-repetition-rate femtosecond laser pulses

Two-stage optical trapping and assembling of protein at air/solution interface

Summary: Optical trapping at interfaces is becoming increasingly important, particularly for proteins with submillimeter structures. Initially, lysozyme clusters are trapped on the surface, gradually forming a thicker layer over time. However, once a concentration threshold is reached, lysozyme clusters in the solution are collected and transported towards the focus, invading the lysozyme layer and creating a border between them, without any detectable concentration jump. This two-stage optical trapping is due to the long-range interaction originating from the focus.

Chiral crystallization manipulated by orbital angular momentum of light

Summary: Researchers have successfully demonstrated the selective control of the chirality of sodium chlorate crystals by altering the helicity of an irradiating optical vortex laser beam. A high crystal enantiomeric excess of 57% is achieved. This study provides fundamental insights into the dynamics and mechanism of enantioselective control in chiral crystallization and the origin of homochirality.

Optical trapping-driven supramolecular photochirogenesis via higher-order complexation

Summary: Despite its potential, the optical trapping method has never been applied to supramolecular chemistry or chiral photochemistry. Here, we demonstrate that optical trapping using a focused laser beam boosts supramolecular assembly beyond the thermodynamic limitations and affords an unprecedented higher-order 2:4 complex through the otherwise-inaccessible dimerization of 1:2 complexes of g-cyclodextrin with two 2-anthracenecarboxylic acids (ACs). This process accelerates the chiral photocyclodimerization of ACs with switched regio-and enantio-selectivities.

Unravelling 3D Dynamics and Hydrodynamics during Incorporation of Dielectric Particles to an Optical Trapping Site

Summary: In this work, the 3D motion trajectories of 200 nm fluorescent polystyrene particles in an optical trapping site were resolved using a widefield multiplane microscope. The particles showed a shallow cone distribution following preferential 3D channels near the focus. The radial and axial flow speed components depended on the axial distance from the focus, which was directly related to the scattering/gradient optical forces. The trapping laser profile determined the particle velocities and trajectories, but collective effects resulting from hydrodynamic forces also played a role.

Unidirectional Optical Swarming of Gold Nanoparticles on Lithographically Fabricated Gold Nanopatterns

These findings provide valuable insights into optical manipulation techniques coupled with fabricated nanostructures, expanding the possibilities for optical trapping and related research.

Effects of plasmon mode on crystallization behaviors by plasmonic trapping

Summary: The study designs and fabricates triangular trimer gold nanostructures with different edge lengths and investigates the effects of plasmon modes on crystallization behaviors. The location of the enhanced electric field is dependent on the edge lengths of the triangular trimer. It is found that polymorphic transformation occurs in the case of continuous laser irradiation on a 230 nm trimer, but not on a 170 nm trimer.
