The Z-configured products are obtained through a base-free CuAAC/ring-opening process due to hydrogen bonding.

Prolinamide promotes novel organocatalytic 1,2-acyl migration and hydrolysis of N-2-carboxyalkylated triazoles in the reaction.

Summary: This study presents a new synthetic strategy for the cycloaddition reaction between non-polarized internal alkynes and azides to yield trisubstituted triazoles. By designing a cyclitol-derived monomer with azide and internal alkyne groups, the reactive orientation necessary for topochemical reaction was achieved through the crystal packing of the monomer molecules. Upon heating, the monomer underwent topochemical polymerization to form a trisubstituted-1,2,3-triazole-linked-polycyclitol.

Summary: A novel triphenylphosphine-mediated [4 + 3] annulation reaction of 2-benzylidene indane-1,3-diones and diynoates was discovered, resulting in biologically interesting indeno[1,2-b]oxepin-4-ylidenes with up to 75% yield. Single-crystal X-ray diffraction confirmed the separable Z and E isomeric oxepins.

Summary: Newly synthesised brominated and phosphonated compounds were investigated for their antiproliferative activities against human cancer cell lines. One of the compounds showed substantial selective cell growth-inhibitory activity against the A2780 cell line and strong interactions with tubulin binding sites. The disturbance of tubulin function was confirmed by photometric polymerisation assay.

The review is divided into two sections based on the number of components in the multicomponent reaction, and provides a critical discussion and analysis of the advantages and disadvantages of each methodology.

Summary: An efficient and simple three-component cascade reaction was reported, providing a variety of polysubstituted imidazo[1,2-a]pyridine derivatives in moderate to excellent yields. The reaction proceeds through a CuAAC/ring-cleavage process and forms a highly active intermediate alpha-acyl-N-sulfonyl ketenimine without the need for a base.
