A potential manufacturing route for the EP1 antagonist GSK269984B was developed. Four synthetic approaches were examined, and a successful realisation of each is presented. The rationale supporting selection of the preferred route is discussed. This route utilised a phenolic aldol reaction as the key step and relied on selective hydrogenolysis to reduce an intermediate diarylmethanol. Further optimisation of the selected route is presented, delivering GSK269984B in three stages and 46% overall yield from readily available starting materials.

A Consideration of the Extent That Tertiary Amines Can Form N-Nitroso Dialkylamines in Pharmaceutical Products

Summary: Most secondary amines can form N-nitrosamines through nitrosation in the presence of nitrite, particularly at low pH. Tertiary amines are less likely to form nitrosamines due to an additional dealkylation step. Simple trialkylamines react slowly to generate N-nitrosamines, making them less of a risk in pharmaceutical products. However, dialkylanilines and structurally complex tertiary amines require a more focused risk assessment due to alternative reaction mechanisms and functional groups that can facilitate nitrosamine formation. Risk assessment should consider the use of simple trialkylamines in situations with higher amounts of nitrosating agents.

Summary: The risk assessment of N-nitrosamines in pharmaceuticals has shifted from focusing on known small-molecule N-nitrosamines to the potential generation of complex nitrosamine drug substance-related impurities. This article proposes a standard set of three orthogonal nitrosation forced degradation types to investigate the potential formation of novel N-nitrosamines from complex amines. The findings from these conditions can be used for risk assessment and further development of synthesis processes for testing purposes.
