An improved synthesis of a selective serotonin reuptake inhibitor

A practical synthesis of 3-((1S, 2S)-2-dimethylaminomethylcyclopropyl)-1H-indole-5-carbonitrile hydrochloride (1), a selective serotonin reuptake inhibitor (SSRI), is described. The process to prepare 1 was demonstrated on laboratory scale and highlights an enantioselective Simmons-Smith cyclopropanation of allylic alcohol 3 using Charette's chiral dioxaborolane ligand. The improved synthesis enabled production of I in 8 chemical steps (5 isolations) in an overall yield of 38%.

A Metal-Free Reductive N-Alkylation of Indoles with Aldehydes

Summary: A metal-free method has been developed for the reductive N-alkylation of indoles using aldehydes and inexpensive Et3SiH as the reductant. A wide range of aromatic and aliphatic aldehydes, as well as various substituted indoles, are viable substrates. The method was successfully demonstrated on a 100 mmol scale for a one-pot sequential 1, 3-alkylation of a substituted indole.

Mechanistic Studies of a Pd-Catalyzed Direct Arylation En Route to Beclabuvir: Dual Role of a Tetramethylammonium Cation and an Unusual Turnover-Limiting Step

Summary: The mechanistic investigation on direct arylation of an indole-containing aryl bromide for the synthesis of the HCV NS5B inhibitor beclabuvir precursor revealed the unique effectiveness of TMAOAc base and an unexpected dual role of tetramethylammonium cation. The study also uncovered an unusual turnover-limiting step in the reaction process. The equilibrium between arylpalladium bromide oxidative addition complex and arylpalladium acetate complex plays a crucial role in the reaction progression, with TMAOAc base driving the reaction forward by sequestering bromide ions as poorly soluble tetramethylammonium salt.

Summary: A new convergent route for the synthesis of NLRP3 agonist BMS-986299 is reported, which involves a key Miyaura borylation and a tandem Suzuki-Miyaura coupling. The synthesis utilizes an iodoimidazole and an o-aminochloroarene as starting materials, leading to the formation of the aminoquinoline core through acid-mediated cyclization. The target compound is obtained through subsequent Boc deprotection and regioselective acylation. This convergent six-step route reduces process mass intensity by 80% compared to the linear enabling synthesis.
