Summary: In this study, it was demonstrated that linear unsymmetrical diaryliodoniums can undergo one-pot cyclization/arylation using a dual nickel/palladium metal system, and the in situ generated benzoxazole motifs can be utilized as directing groups for subsequent C-H activated arylation from the unwanted aryl iodide, exhibiting good atom economy and regioselectivity.

Summary: This study investigates the regioselectivity of C-H amination reactions using hydroxylamine-derived electrophilic aminating reagents on diversely substituted arenes. Summary: A transition metal-free approach using unsymmetric diaryliodonium salts with anisyl ligands has been developed for the N-arylation of amino acid derivatives, achieving high chemoselectivity and yields. This method is applicable for transferring electron deficient, electron rich, and sterically hindered aryl groups, and a cyclic diaryliodonium salt was successfully employed. The N-arylated products were obtained with retained enantiomeric excess.

Summary: A method using pyridone as a directing group to functionalize the arene ring of pirfenidone has been demonstrated. The alkenylation of the N-aryl ring of pirfenidone with internal alkynes using a ruthenium catalyst enables the synthesis of new drug analogues in a step-economical manner, due to its high functional group tolerance and site-selective functionalization.

Summary: An efficient method for the synthesis of multi-substituted cyclic imides was developed using cyanoesters and diaryliodonium salts. This method involves a cascade of N-arylation-acylation and rearrangement reactions, producing the desired heterocycles in high yields. The method has a broad substrate scope, excellent functional group compatibility, and can be extended to fused cyclic imides.

Summary: A two-step strategy using unsymmetrical iodonium salts as versatile synthetic linchpins for transition-metal-free C-H functionalization of arenes is presented. The success relies on the identification of the 3,5-dimethyl-4-isoxazolyl (DMIX) group as a superior dummy ligand, enabling site-selective C-H functionalization and highly selective aryl transfer. Diverse structural elaboration is achieved through metal-free C-N, C-C, C-S, and C-O coupling.

Summary: By utilizing a chiral directing group, asymmetric rhodium(III)-catalyzed C-H bond addition to aldimines followed by intramolecular cyclization to form chiral isoindolinones has been achieved, with up to 68% yield and up to 93% ee. A three-component variant resembling Mannich reaction was also successful, with a yield of 41% and 83% ee. The study also includes product elaborations and preliminary mechanistic studies.

Summary: A mild and efficient copper-mediated trifluoromethylation method was developed for the synthesis of trifluoromethylated arenes using readily available fluoroform and diaryliodonium salts. The reaction showed excellent functional group tolerance and provided good yields. This approach enables the straightforward incorporation of CF3 group into complex molecules, which is important for drug discovery applications.

Summary: Arylation of various phosphines with aryl(mesityl)iodonium triflates under blue light irradiation, followed by oxidative P-N bond cleavage in the in situ generated amino phosphonium salts under hydrolytic conditions, allows for the synthesis of substituted arylphosphine oxides, arylphosphinic, and arylphosphonic amides. This method utilizes visible light as the sole promoter for C-P bond formation, is compatible with a wide range of functional groups, and can be applied for late-stage C-H functionalization of drug molecules.

By utilizing the cyclization reaction between sulfinylidene benzamides and vinylene carbonate, along with directing group-assisted ortho-C-H coupling and intramolecular alcoholysis, nonsubstituted isocoumarins can be efficiently synthesized.

Summary: A [2 + 2 + 1] annulation protocol has been developed for the modular synthesis of 2,4-disubstituted thiophenes/selenophenes, showing excellent regioselectivity. Catalyzed by copper salt, the reactions utilize elemental sulfur and selenium as the chalcogen source. Mechanistic studies indicate that the process is initiated by a trisulfur radical anion detected using EPR.

Summary: Introducing the primary amino group into aromatic ring is a crucial task in organic chemistry, especially in the field of pharmaceuticals and agrochemicals. In this study, we present a photoinduced direct C-H amination of arenes with ammonia using site-selective C-H thianthrenation, which leads to excellent regioselectivity. The reaction conditions are mild and a wide range of functional groups, such as sensitive -Cl, -Br, and -OH groups, can be tolerated, which is challenging in conventional approaches. Moreover, the synthetic utility of our amination protocol has been demonstrated through late-stage modification and gram-scale synthesis of complex drug-like molecules.

Summary: Here we describe a manganese-catalyzed hydroalkenylation reaction that selectively introduces branched groups to terminal alkynes under mild conditions using a removable silanol directing group. Coupling with an alkenyl boronic acid partner yields (E,E)-1,3-dienes with high regio-, chemo- and stereoselectivity. The reaction proceeds at room temperature in air and is compatible with various functional groups. The resulting 1,3-dienesilanol products can be used as versatile building blocks for downstream synthesis of branched or linear 1,3-dienes and 1,3,5-trienes by removing the silanol group. Additionally, a Diels-Alder cycloaddition delivers silicon-containing pentasubstituted cyclohexenes. Mechanistic studies and DFT calculations support a bimetallic synergistic activation model for the enhanced catalytic efficiency and regioselectivity observed.

Summary: This study reports a copper-catalyzed ring opening reaction of carbon dioxide (CO2) with amines, leading to the synthesis of a range of axially chiral carbamates. The method offers mild conditions, broad substrate scope, and excellent enantioselectivity. The synthetic applications of the method were demonstrated through the modification of natural products and drug molecules, as well as the diverse transformations of the axially chiral products.
