Summary: Breast cancer is the most frequently diagnosed cancer among women worldwide, and elevated cytoplasmic p21 levels are associated with poor prognosis. The underlying mechanisms that lead to the stabilization of cytoplasmic p21 protein, which normally has a short half-life, remain unclear. In this study, we found a strong correlation between p21 and USP11 in the cytoplasm of breast cancer tissues and cells. We also revealed that ERK1/2 phosphorylated USP11 at the Ser905 site, promoting its cytoplasmic localization. The colocalization and interaction between USP11 and p21 in the cytoplasm resulted in the removal of polyubiquitin chains from cytoplasmic p21, thereby stabilizing it. Functionally, the USP11-mediated stabilization of cytoplasmic p21 induced breast cancer cell proliferation both in vitro and in vivo. Our findings provide evidence that ubiquitinated p21 in the cytoplasm can be recycled through USP11-mediated deubiquitination, and suggest the USP11-p21 axis as a potential therapeutic target for breast cancer control.

Inhibition of p53 expression reduced the number of senescent myoblasts, indicating that sepsis-induced cellular senescence is dependent on p53. Additionally, metformin attenuated muscle senescence and improved muscle strength in septic mice, suggesting it as a potential therapeutic drug for ICU-AW.

Summary: This study aimed to evaluate the inhibitory effect of nobiletin on breast cancer and elucidate potential mechanisms against invasion and migration. The results showed that nobiletin can inhibit the invasion and migration of breast cancer cells by suppressing the ERK and JNK signaling pathways. In addition, nobiletin inhibited liver metastasis of breast cancer in vivo. Therefore, nobiletin has the potential to be used as an anticancer agent.
