Summary: This study reveals that the Erk1/2 inactivation-triggered and COP1-mediated c-Jun degradation is regulated by protein phosphatases, UBE2 enzymes, and an intrinsic motif of c-Jun. The C-terminus of c-Jun protein plays a critical role in facilitating its degradation.

Summary: This study identified a key molecular mechanism that influences lymph node metastasis. RPRD1B was found to promote lymph node metastasis by regulating the c-Jun/c-Fos/SREBP1 axis, and NEAT1 enhanced the activity of this axis through a positive feedback loop. These findings provide a potential therapeutic target for the treatment of gastric cancer.

Summary: This study investigates the role of TEX11 gene in colorectal cancer (CRC). The results show that TEX11 expression is significantly downregulated in clinical samples and cell lines, and is associated with poor prognosis in CRC patients. Further experiments demonstrate that TEX11 can inhibit the proliferative capacity of CRC cells. Mechanistically, TEX11 promotes COP1 transcription by upregulating FOXO3a expression, thus accelerating the degradation of the negative transcriptional regulator c-Jun and enhancing p21 transcription, leading to inhibition of CRC cell proliferation.

Summary: This study demonstrates that TRIB2 reduces Ub levels by stimulating proteasome degradation and interacts with PCBP2 to modulate PSMB5 activity, which is crucial for liver cancer cell viability and tumor growth. The interaction between TRIB2 and PCBP2 also protects liver cancer cells from oxidative damage through the function of glutathione peroxidase 4. Targeting TRIB2 may enhance the effectiveness of therapeutic agents for liver cancer treatment.

Summary: This study reveals that UTX promotes colorectal cancer (CRC) progression through downregulation mediated by the CUL4B-DDB1-COP1 complex. Utx deletion in mice increases susceptibility to tumorigenesis, but this effect can be alleviated by the EZH2 inhibitor GSK126. Furthermore, the CUL4B-DDB1-COP1 complex is identified as the functional E3 ligase responsible for targeting UTX for degradation in CRC cells. Patient cohort analysis also shows that UTX expression is negatively correlated with clinical stage, favorable disease outcomes, and COP1 expression.

Summary: The study reveals that KAT6A is upregulated in ovarian cancer and correlates with patient survival. Downregulation of KAT6A inhibits proliferation and migration of ovarian cancer cells, induces apoptosis, and enhances sensitivity to cisplatin. Additionally, KAT6A interaction with COP1 leads to impaired function, accumulation, and enhanced activity of beta-catenin, suggesting a critical role of the KAT6A/COP1/beta-catenin signaling axis in ovarian cancer progression. Targeting this signaling axis could be a novel strategy for ovarian cancer treatment.

Summary: S6K1 plays a crucial role in regulating cell proliferation and growth in the mTOR signaling pathway. Our study demonstrates the dynamic distribution of S6K1 in the cytoplasm and nuclei of human cervical cancer cells and reveals its additional role as a gene transcription regulator. These findings provide important insights into the mechanisms of tumorigenesis and potential therapeutic strategies.

Summary: This study revealed the significant role of copy number aberrations in cancer evolution, as well as the importance of neoantigens and diverse T cell receptor repertoire in postoperative recurrence. The comparison of driver mutations and immune response-related molecules between early and metastatic sites showed differences, and the postoperative recurrence was associated with neoantigen levels and TCR diversity in the metastatic sites.

Summary: The detection and sequencing of mutated ctDNA is essential in postoperative management of colorectal cancer (CRC). This study conducted target resequencing of ctDNA and found that mutated genes in ctDNA indicated immune-resistance traits due to the loss of neoantigen presentation in primary CRC tumors. Therefore, ctDNA mutations can be frequently detected postoperatively to identify recurrence, but they come from immune-tolerated clones.

Summary: Gastric adenocarcinoma and proximal polyposis of the stomach (GAPPS) is a rare familial gastric cancer syndrome caused by a mutation in the APC gene. Treatment involves standard methods for gastric cancer, with prognosis depending on the presence of distant metastasis and the use of total gastrectomy. Prophylactic total gastrectomy is recommended in cases of dysplasia, but careful consideration of the patient's background is necessary. Currently, there are no guidelines for screening, timing of prophylactic total gastrectomy, or endoscopic surveillance for GAPPS.
