Summary: Cancer is a genetic disease caused by changes in genes controlling cell growth. The Hippo pathway, a developmental signaling system, plays a critical role in tissue and organ size regulation and is frequently deregulated in cancer. This article reviews the relevance of the Hippo pathway in specific sarcoma subtypes and discusses its genetic alterations and other molecular mechanisms involved in its deregulation. It also explores the potential for targeted therapy in sarcoma based on these alterations.

Summary: This study demonstrates that Bufalin can inhibit the proliferation of lung cancer cells in vivo/in vitro by suppressing the Hippo-YAP pathway. The findings show that Bufalin promotes the binding of LATS and YAP, leading to increased YAP phosphorylation. Phosphorylated YAP is unable to enter the nucleus and activate the expression of downstream proliferation-related genes, while the retained YAP in the cytoplasm undergoes degradation. This study verifies the key role of YAP in stimulating lung cancer proliferation and identifies Bufalin as a potential anticancer drug target.

Summary: The Hippo signaling pathway regulates various cellular processes, including cell proliferation, differentiation, migration, and mechanosensation. Calcium ion (Ca2+) has been found to influence core components of the Hippo pathway. However, the crosstalk between Ca2+ signaling and the Hippo pathway is not fully understood.

Summary: Epithelioid hemangioendothelioma (EHE) is a rare vascular cancer with a unique WWTR1(TAZ)-CAMTA1 gene fusion. Studying this disease can provide insights into how dysregulation of TAZ promotes tumorigenesis and these findings can be applied to other YAP/TAZ-dependent cancers.

Summary: The aim of this study was to investigate the mechanism of interaction between myocardial ischemia-reperfusion (I/R) injury and Resveratrol (Res). The results showed that Resveratrol could ameliorate necroptosis induced by myocardial I/R injury by modulating the Hippo pathway, and the beneficial effect might be associated with nuclear translocation of the transcriptional regulator YAP.
