Summary: Nasopharyngeal carcinoma (NPC) is a highly diverse cancer associated with oncogenic Epstein-Barr Virus (EBV), characterized by late-stage diagnosis and low survival rate. The current radiotherapy and chemotherapy have limited effectiveness and adverse effects, necessitating the development of new therapies. This study demonstrates that adoptive immunotherapy using gamma delta T cells, combined with butyrophilin 2A1 and 3A1 protein expression, shows promise as a potential treatment for NPC.

Summary: Nasopharyngeal carcinoma is a malignancy associated with Epstein-Barr virus and epigenetic abnormalities play important roles in its development. The latent membrane protein 1 (LMP1) is a key factor in inducing aberrant DNA methylation of tumour suppressor genes. This study identified LMP1-inducible methylation genes and demonstrated that DERL3 repression by DNA methylation contributes to the progression of nasopharyngeal carcinoma. The findings suggest potential targets for therapeutic intervention.

Summary: The Asia Pacific EBV strain Yu103 exhibits a unique capacity to infect and transform both B-lymphocytes and nasopharyngeal epithelial cells, lying midway along the spectrum of EBV strains known to drive lymphomagenesis or carcinogenesis. This EBV isolate is currently the only one shown to drive human nasopharyngeal carcinoma and B-lymphoma, making it a powerful novel platform for research on EBV-driven hematological and epithelial malignancies.

Summary: Bmi-1 is overexpressed in various cancer types, including nasopharyngeal carcinoma (NPC). High Bmi-1 levels were observed in NPC cell lines and specimens, especially in advanced cases. Depletion of Bmi-1 suppressed cell proliferation, induced cell cycle arrest, reduced cell stemness, and inhibited cell migration and invasion in NPC cells. Hairy gene homolog (HRY) upregulated Bmi-1 by binding to its promoter, promoting NPC cell stemness. Silencing Bmi-1 suppressed NPC progression.

Summary: This study reveals the interaction between TNF alpha released by macrophages and EBV-encoded LMP1 in driving the mobilization of invadopodia in EBV-associated nasopharyngeal carcinoma (NPC). The mobilizing invadopodia exhibit lateral movements and enhanced degradative power, transforming from dot-like digestion patterns to dendrite-like patterns. EBV infection sensitizes NPC cells to form mobilizing invadopodia in a TNF alpha-rich tumor microenvironment.

Summary: In this study, it was found that NPC cell-derived exosomes containing LMP1 promoted proliferation, migration, tubulogenesis, and permeability in ECs through the Akt/ERK pathway. Silencing STIM1 reduced LMP1 enrichment in NPC cell-derived exosomes and reversed its pro-oncogenic effects. Furthermore, STIM1 knockdown in NPC cells inhibited tumor-induced vascular network formation and intra-tumor neovascularization.
