Mitochondrial sirtuins in stem cells and cancer

Amit Jaiswal, Zhu Xudong, Ju Zhenyu, Gabriele Saretzki

Summary: Mitochondrial sirtuins, part of the mammalian sirtuin family, play crucial roles in regulating mitochondrial function and cell fate by interacting accurately with delicate processes mediated by crosstalk between the nucleus and mitochondria. While previous studies have provided ample information on their substrates and targets, their role in cancer and stem cells remains less understood, with recent advances shedding light on their structural and functional properties in these contexts.

WHO essential medicines for children 2011-2019: age-appropriateness of enteral formulations

Ebiowei Samuel F. Orubu, Jennifer Duncan, Catherine Tuleu, Mark A. Turner, Anthony Nunn

Summary: The study found that enteral formulations in the WHO Essential Medicine List for children were more age-appropriate for older children (6-11 years old) than for younger children. Only a small proportion of formulations were considered age-appropriate for children under 6 years old. However, medications for targeted diseases like HIV and tuberculosis were mostly age-appropriate in the list.

Summary: Neonatal sepsis, a leading cause of infant mortality, has diverse definitions based on microbiological culture, laboratory tests, and clinical signs. An international consensus-based definition could improve research comparability and clinical outcomes.

Miscarriage syndrome: Linking early pregnancy loss to obstetric and age-related disorders

Pietro Bortoletto, Emma S. Lucas, Pedro Melo, Ioannis D. Gallos, Adam J. Devall, Tom Bourne, Siobhan Quenby, Phillip R. Bennett, Arri Coomarasamy, Jan J. Brosens

Summary: After embryo implantation, the endometrium transforms into a robust decidual matrix to support the fetal placenta. Circulating NK cells and BM-MSC play a crucial role in decidual homeostasis and expansion in early pregnancy. Pathological signals that interfere with the recruitment or activity of extrauterine cells at the maternal-fetal interface may be linked to miscarriage and subsequent adverse pregnancy outcomes.

Ethnic group and reason for assisted reproductive technology failure analysis of the Human Fertilisation and Embryology Authority registry data from 2017 to 2018

Ian Henderson, Lauren Lacey, Muhammad Ahsan Akhtar, Siobhan Quenby

Summary: This study aimed to understand the risk differences of different assisted reproductive technology (ART) failure types by ethnic group and explore the role of maternal age and suspected etiology as mediators. The study conducted an observational analysis of 48,750 women who underwent ART treatment in the United Kingdom. The results showed that black women had the highest risk of all failure types, while Asian women had an intermediate risk compared to white women. Some of the risks among black women may be mediated by maternal age.

The publication quality of laboratory values in clinical studies in neonates

Karel Allegaert, Heidrun Hildebrand, Kanwaljit Singh, Mark A. A. Turner

Summary: The International Neonatal Consortium aims to define reference ranges for commonly used laboratory values in neonates, but there is a lack of standardized and complete published information on these values. This hinders treatment comparison, safety monitoring, and clinical management.

The Telomerase Connection of the Brain and Its Implications for Neurodegenerative Diseases

Gabriele Saretzki

Summary: Telomerase, consisting of TERT and TERC, is well-known for maintaining and extending telomeres. However, TERT has non-canonical functions, including shuttling to the cytoplasm and mitochondria to protect cells from stress. TERT also plays a protective role in brain neurons by countering effects of toxic proteins and activating protein degradation pathways, suggesting its potential in treating neurodegenerative diseases.

Summary: The objective of this study was to develop core outcome sets (COS) for miscarriage management and prevention. Through a modified Delphi survey and consensus development meeting, stakeholders from 15 countries were included. The final COS for miscarriage management and prevention were established, providing important guidance for future randomized trials and systematic reviews.

Fertility preservation, its effectiveness and its impact on disease status in pre-menopausal women with breast cancer: A systematic review and meta-analysis

Rhiannon White, Anna Wilson, Natasha Bechman, Stephen D. Keay, Lucy McAvan, Siobhan Quenby, Joshua Odendaal

Summary: This systematic review aimed to investigate the effectiveness and safety of fertility preservation strategies in pre-menopausal women with breast cancer. The results showed that fertility preservation interventions were associated with higher fertility outcomes and a reduced rate of disease recurrence. This suggests that fertility preservation is effective in preserving reproductive function and safe with regard to disease recurrence, disease free survival, and overall survival in premenopausal women with breast cancer.
