Summary: This study found that heterozygous variants in LEP, LEPR, POMC, and PCSK1 are common in severe obesity and sometimes exhibit a phenotype similar to homozygotes. Summary: The study investigated the dopamine receptors expression in adipose tissue of patients with T2D and the therapeutic effects of bromocriptine in modulating metabolism in this tissue. Patients with insulin resistance showed decreased DRD1 and DRD4 expression, while bromocriptine treatment in diabetic rats increased D1R and TH levels, leading to higher insulin sensitivity and improved metabolic status. Bromocriptine treatment remodeled the adipose tissue and liver dopaminergic system, resulting in enhanced insulin sensitivity and catabolic function.

Summary: This study evaluated the obesogenic effect of 2-ethylhexyl diphenyl phosphate (EHDPHP) on zebrafish. Results showed that EHDPHP exposure promoted feeding behavior and caused obesity in zebrafish. The interaction between EHDPHP and the dopamine receptor D2 (DRD2) was identified through molecular docking. Furthermore, EHDPHP disrupted pathways related to neurological disorders.

Summary: The difference in the secondary binding site between D2R and D3R has been utilized to design compounds with selectivity for D3R. This study prepared a series of bitopic ligands based on Fallypride to improve the selectivity for D3R using various secondary binding fragments. The results showed that compounds with a small alkyl group containing a heteroatom exhibited improved D3R selectivity.
