Serum Amyloid A Induces Lipolysis by Downregulating Perilipin Through ERK1/2 and PKA Signaling Pathways

Serum amyloid A (SAA) is not only an apolipoprotein, but also a member of the adipokine family with potential to enhance lipolysis. The purpose of this study was to explore how SAA facilitates lipolysis in porcine adipocytes. We found that SAA increased the phosphorylation of perilipin and hormone-sensitive lipase (HSL) after 12-h treatment and decreased perilipin expression after 24-h treatment, and these effects were prevented by extracellular signal-regulated kinase (ERK) or protein kinase A (PKA) inhibitors in primary adipocyte cell culture. SAA treatment decreased HSL and adipose triglyceride lipase (ATGL) expression. SAA treatment also activated ERK and PKA by increasing the phosphorylation of these kinases. Moreover, SAA significantly increased porcine adipocyte glycerol release and lipase activity, which was inhibited by either ERK (PD98059) or PKA (H89) inhibitors, suggesting that ERK and PKA were involved in mediating SAA enhanced lipolysis. SAA downregulated the expression of peroxisome proliferator-activated receptor gamma (PPAR gamma) mRNA, which was reversed by the ERK inhibitor. We performed a porcine perilipin promoter assay in differentiated 3T3-L1 adipocytes and found that SAA reduced the porcine perilipin promoter specifically through the function of its PPAR response element (PPRE), and this effect was reversed by the ERK inhibitor. These findings demonstrate that SAA-induced lipolysis is a result of downregulation of perilipin and activation of HSL via ERK/PPAR gamma. and PKA signaling pathways. The finding could lead to developing new strategies for reducing human obesity.

Cryptic Diversity, Molecular Systematics, and Pathogenicity of Genus Pestalotiopsis and Allied Genera Causing Gray Blight Disease of Tea in Taiwan, With a Description of a New Pseudopestalotiopsis Species

Summary: This study identified and characterized the causative agents of tea gray blight disease in Taiwan, revealing seven well-classified taxa and seven tentative clades within three genera, including one novel species and five newly recorded species. Pathogenicity assessments showed that all assayed isolates were pathogenic on tea leaves, with Pseudopestalotiopsis chinensis and Pseudopestalotiopsis camelliae-sinensis identified as the major pathogens associated with gray blight disease of tea in Taiwan. This study is the first to focus on the diversity, pathogenicity, and characterization of pestalotiopsis-like fungi causing tea gray blight disease in Taiwan.

Summary: This study reveals that lysophosphatidic acid (LPA) has opposing regulatory effects on erythrocyte differentiation through activation of LPA receptors 2 and 3. The expression of LPA(2) and LPA(3) in murine myeloid cells may determine the direction of myeloid differentiation, with LPA(2) inhibiting early myeloid differentiation and LPA(3) promoting erythrocyte production. Activation of LPA(3) is essential for the restoration of acute hemolytic anemia and correlates with erythropoiesis impairment in a premature aging model.

The Role of Small Extracellular Vesicles in the Progression of Colorectal Cancer and Its Clinical Applications

Neutralization or enhancement of SARS-CoV-2 infection by a monoclonal antibody targeting a specific epitope in the spike receptor-binding domain

Summary: Two mouse monoclonal antibodies, 7 Eb-4G and 1Ba-3H, were identified to specifically recognize the receptor-binding domain (RBD) of SARS-CoV-2 spike protein. Only 1Ba-3H exhibited neutralizing activity and interfered with the binding between RBD and ACE2. Epitope mapping experiments revealed specific residues essential for 1Ba-3H binding. Interestingly, 1Ba-3H showed neutralizing activity against several SARS-CoV-2 variants but enhanced cellular infection of the Gamma variant.

Autologous Platelet-Rich Growth Factor Reduces M1 Macrophages and Modulates Inflammatory Microenvironments to Promote Sciatic Nerve Regeneration

Summary: The failure of peripheral nerve regeneration may be attributed to the lack of a permissive environment for nerve repair. The use of autologous platelet-rich plasma (PRP) or its derivative platelet-rich growth factors (PRGF) has been shown to improve peripheral nerve regeneration by modulating macrophage polarization. In this study, PRGF was found to reduce inflammation and promote the shift of macrophages from a pro-inflammatory M1 phenotype to an anti-inflammatory M2 phenotype. This led to improved nerve regeneration in a rat sciatic nerve transection model. The findings suggest that autologous PRGF has the potential to regulate inflammatory microenvironments and promote nerve repair.

Analysis of the population genetic structure using microsatellite markers in goat populations in Taiwan
