Disintegration of wheat aleurone structure has an impact on the bioavailability of phenolic compounds and other phytochemicals as evidenced by altered urinary metabolite profile of diet-induced obese mice

Background: Phenolic acids are covalently bound to the arabinoxylan fibre matrix of wheat aleurone layer. In order to be bioavailable they need to be released by endogenous or bacterial enzymes and absorbed within the intestinal lumen. The intestinal microbiota can metabolize phenolic acids and other food-born phytochemicals. However, the effect of structure of the cereal bran or aleurone layer on these processes is not comprehensively studied. Methods: The structure of aleurone layer was modified either by dry-grinding or by enzymatic treatments with xylanase alone or in combination with feruloyl esterase. Diet induced obese C57BL6/J mice were fed with high-fat diets containing either pure ferulic acid, or one of the four differentially treated aleurone preparations for 8 weeks. The diets were designed to be isocaloric and to have similar macronutrient composition. The urinary metabolite profiles were investigated using non-targeted LC-qTOF-MS-metabolomics approach. Results: The different dietary groups were clearly separated in the principal component analysis. Enzymatic processing of aleurone caused increased excretion of ferulic acid sulfate and glycine conjugates reflecting the increase in unbound form of readily soluble ferulic acid in the diet. The urinary metabolite profile of the diet groups containing native and cryo-ground aleurone was more intense with metabolites derived from microbial processing including hippuric acid, hydroxyl-and dihydroxyphenylpropionic acids. Furthermore, aleurone induced specific fingerprint on the urinary metabolite profile seen as excretion of benzoxazinoid metabolites, several small dicarboyxlic acids, and various small nitrogen containing compounds. Conclusions: The structural modifications on wheat aleurone fraction resulted in altered metabolism of aleurone derived phenolic acids and other phytochemicals excreted in urine of diet-induced obese mice.

Diet- and microbiota-related metabolite 5-aminovaleric acid betaine (5-AVAB), in health and disease

Summary: 5-Aminovaleric acid betaine (5-AVAB) is a gut microbiota-associated compound that accumulates in metabolically active tissues. It has been found in higher concentrations in the heart, muscle, and brown adipose tissue. It has positive health effects on fetal brain development, insulin secretion, and reduced cancer risk, but it is also associated with negative health outcomes such as cardiovascular disease and fatty liver disease.

An inverse association between plasma benzoxazinoid metabolites and PSA after rye intake in men with prostate cancer revealed with a new method

Summary: In this study, a quantitative method for analysis of benzoxazinoids (BXs) was established, and the plasma levels of BXs after consumption of whole grain/bran rye or refined wheat were investigated in men with prostate cancer. The results showed that BX metabolites were significantly higher after rye consumption compared to refined wheat. Additionally, four BX-related metabolites were inversely associated with PSA levels in plasma. These findings suggest that rye may have potential preventive effects against prostate cancer.

Effects of exercise on NAFLD using non-targeted metabolomics in adipose tissue, plasma, urine, and stool

Summary: This study investigated the metabolic changes associated with non-alcoholic fatty liver disease (NAFLD) in response to high-intensity interval training (HIIT) intervention. The results showed that HIIT significantly improved clinical parameters of NAFLD patients, including fasting plasma glucose concentration and waist circumference. Metabolomics analysis revealed sample-type-specific changes in metabolite levels, with amino acids and their derivatives accumulating in adipose tissue and plasma, while decreasing in urine and stool. The study highlights the role of adipose tissue in responding to HIIT and suggests that altered amino acid metabolism in adipose tissue may play a critical role in improving fasting plasma glucose concentration.

Associations of PFAS-related plasma metabolites with cholesterol and triglyceride concentrations

Summary: The widespread environmental pollutants PFAS have been repeatedly linked to elevated serum cholesterol in humans, but the underlying mechanisms are still unclear. In this study, we investigated the associations between PFAS-related metabolite features and plasma cholesterol and triglyceride concentrations. The results indicated that longer chain PFAS may affect glycerophospholipid metabolism, leading to changes in triglyceride levels.

Associations of altered hepatic gene expression in American lifestyle-induced obesity syndrome diet-fed mice with metabolic changes during NAFLD development and progression

Summary: This study aimed to elucidate the pathogenesis of NAFLD and its relationship with metabolic alterations. ALIOS diet-fed mice showed higher hepatic steatosis, body weight, energy consumption, and liver mass compared to control mice. The ALIOS diet also altered the expression of genes related to inflammation and lipid metabolism. Metabolomics analysis revealed changes in lipid and peptide profiles, as well as correlations between different metabolites and inflammation, lipid uptake, and synthesis. The reduction of antioxidant metabolites and gut microbiota-derived metabolites contributed to NAFLD development. Non-targeted metabolomics combined with gene expression can help identify potential therapeutic targets for NAFLD.

Several microbial metabolites have been identified in this context, with many important metabolites yet to be discovered.
