Summary: We present a short synthesis method for novel C-nucleoside Remdesivir analogues and evaluate their cytotoxicity and in vitro efficacy against SARS-CoV-2 (CoV2). These compounds are nucleoside analogues with a nitrogen heterocycle as purine analogues. The designed hybrid structures aim to enhance the anti-CoV2 activity of Remdesivir. We discuss the impact of sugar and base modifications on the biological activities of these compounds, their lack of cytotoxicity, and their antiviral efficacy.

Summary: The study aimed to present an overview of NAs-based drug repurposing for targeting SARS-CoV-2 RdRp through computational analysis. Additionally, the clinical development of some NAs including Favipiravir, Sofosbuvir, Ribavirin, Tenofovir, and Remdesivir as potential inhibitors of RdRp was surveyed. Overall, exploring broad-spectrum NAs as promising inhibitors of RdRp may provide useful information about potential antiviral repurposed drugs against SARS-CoV-2.

Summary: Different viruses, such as enterovirus 71, rely on the host enzyme METTL3 to complete their cytoplasmic life cycle stages. By modulating the activity of this enzyme, it is possible to interfere with a broad range of viral infections. In this study, a series of nucleoside analogues were designed as inhibitors of human METTL3 to target multiple viral infections. Through molecular docking studies and synthesis, several novel and potent inhibitors of enterovirus 71 were identified.
