Summary: In this study, we conducted a genome-wide identification, evolutionary relationship analysis and gene expression analysis of the HSP70, HSP90, and HSF gene families in Ruditapes philippinarum. We identified 83 RpHSP70, 6 RpHSP90, and 3 RpHSF genes in R. philippinarum. The structural characteristics, chromosomal localization, and the gene structure map were constructed to reveal the characteristics of protein structures. Furthermore, the expression profiling of transcriptome data showed the expression pattern of HSP70, HSP90 and HSF genes in Manila clam from different populations, and under high and low temperature stress. In addition, we performed protein-protein interaction network analysis between HSP70, HSP90, and HSF gene family which enabled us to recognize the regulatory relationship between the two HSP gene families and the HSF gene family. Furthermore, the predicted sub-cellular location revealed a diversified subcellular distribution of HSP70, HSP90, and HSF proteins, which may be directly or indirectly associated with functional diversification under heat stress condition.

Summary: This study aimed to investigate the inhibitory effect of methanolic extracts from Pulsatilla vulgaris Mill. on HeLa cell proliferation by modulating cancer-related signaling pathways. The study also determined the cytotoxicity and chemical composition of P. vulgaris extracts from leaves and roots. The results showed that the root extract of P. vulgaris inhibited 12 signaling pathways in cervical cancer cells, with the most significant inhibition observed for MYC, Notch, Wnt, E2F, Ets, Stat3, Smad, Hdghog, AP-1, and NF-kappa B at a concentration of 40 μg/mL. The methanolic extracts of P. vulgaris induced apoptotic death and disrupted cellular proliferation, differentiation, and progression towards a neoplastic phenotype by altering key signaling molecules necessary for cell cycle progression. Additionally, phytochemical analysis revealed that compounds strongly suppressing the growth and proliferation of HeLa cancer cells in the methanolic extracts of P. vulgaris were mainly triterpenoid saponins accompanied by phenolic acids.

Summary: Loss of olfactory function in SARS-CoV-2 infections is associated with sustentacular cell infection and inflammation. Infection leads to damage and loss of cilia on olfactory sensory neurons (OSNs) and downregulation of olfactory receptors and signal transduction molecules. Recovery of olfactory function takes about 3-4 weeks, and not all animals fully recovered. Similar patterns of olfactory infection and dysfunction were observed in mice infected with other human coronaviruses.

Summary: The authors demonstrate that the human chaperone system consists of ubiquitous core chaperones and tissue-specific variable chaperones, and perturbation of which leads to tissue-specific phenotypes. The core chaperones are significantly more abundant across tissues and important for cell survival, forming tissue-specific functional networks. This functional organization is established in development and declines with age, highlighting the importance of understanding tissue-specific differences in protein folding capacities.
