Summary: This study reveals the dysfunction of RNA binding protein hnRNP A1 in oligodendrocytes (OLs) in patients with multiple sclerosis (MS) as well as in an animal model of MS. The dysfunction of hnRNP A1 is associated with alterations in RNA metabolism, cell morphology, and programmed cell death pathways in OLs, leading to detrimental effects on OL functioning and survival. These findings suggest a potential mechanism of OL damage and death in MS.

Summary: The Caenorhabditis elegans model has provided insights into the role of G-quadruplexes in genomic instability. Telomeric sequences in C. elegans show similarities to those in humans, but even small changes in these sequences can have significant effects on the folded G-quadruplex structure. This study determined the structure of the C. elegans telomeric sequence, revealing an intramolecular two G-tetrads basket type G-quadruplex structure with a C-T base pair in the diagonal loop. The findings shed light on the telomeric structure of the widely used C. elegans animal model.

Summary: We investigated the stacking interactions and structural features of telomeric G4 multimers using a low-resolution structural approach that combines small-angle X-ray scattering (SAXS) with extremely coarse-grained (ECG) simulations. The self-assembly of G4 multimers leads to significant polydispersity and exponential distribution of contour lengths. We found that the strength of the stacking interaction between G4 monomers increases with increasing DNA concentration, as well as the average number of units in the aggregates. The proposed methodology may aid in the selection and design of drugs that target G4s under physiological conditions.

Summary: The multifunctional adaptor protein PCBP1 coordinates single-stranded nucleic acids and iron-glutathione complexes, with separate activities controlling cell cycle progression and DNA damage suppression while maintaining cell viability. Amino acid residues required for iron coordination on each structural domain of PCBP1 were identified, confirming their essential function in binding target proteins. The coevolution of RNA/DNA binding and iron chaperone activities provides an advantage for nucleic acid processing dependent on iron cofactors.

Summary: Telomeres are part of a highly refined system for maintaining the stability of linear chromosomes, with most relying on repetitive sequences and telomerase enzymes. However, in some cases, an alternative lengthening of telomeres (ALT) mechanism is used, which mainly utilises recombination-based replication mechanisms. The study on mouse telomeres reveals the use of non-telomeric sequences and a specific subtelomeric element called mTALT for repairing telomeric DNA damage and composing portions of ALT-dependent telomeres in mouse embryonic stem cells.
