Summary: This study demonstrated that ginsenoside Rg3 reversed the TNF-α-inhibited SIRT1 expression, and suppressed TNF-α-induced CypD acetylation by activating the SIRT1/PGC-1 alpha/SIRT3 pathway, leading to reduced mitochondrial dysfunction and ROS accumulation. Additionally, Rg3 reduced ROS levels, ameliorated TNF-α-induced apoptosis, and mediated the downregulation of p38 MAPK to inhibit NF-kappa B translocation. Overall, Rg3 may serve as an adjunct therapy for arthritis patients by regulating the SIRT1/PGC-1 alpha/SIRT3/p38 MAPK/NF-kappa B signaling pathway.

Summary: This study compares preserved and damaged cartilage in patients with osteoarthritis (OA) and identifies a significant reduction of estrogen receptor alpha (ER alpha) in damaged cartilage. Further investigations reveal that ER alpha plays an important role in mediating chondrocyte phenotype and response to mechanical loading. Inhibiting ER alpha expression enhances chondrocyte senescence and osteoarthritic phenotype. These findings suggest that enhancing ER alpha level may represent a new method to treat osteoarthritis.

Summary: This study investigates the impact of SIRT1 on the anti-HBV activity of IFN-alpha and its underlying mechanism. The results show that silencing SIRT1 enhances the antiviral effects of IFN-alpha against HBV replication by activating the JAK-STAT pathway and inhibiting Pol expression. Additionally, SIRT1 interacts with Pol to regulate JAK-STAT activity. These findings suggest that targeting SIRT1 could improve the efficacy of IFN-alpha in the treatment of chronic hepatitis B.

Summary: CRISPR-Cas9 is a powerful tool for genome engineering, and a LacI-reporter system was developed in the study to evaluate gRNA cleavage efficiency by quantitating luciferase activity or EGFP expression. The study found a strong positive correlation between the efficiency of gRNAs measured using this system and mutation frequency measured using surveyor and deep sequencing, with validation in human liver organoids. The LacI-reporter system provides a useful tool for selecting efficient gRNAs for genome editing.
