UCbase & miRfunc: a database of ultraconserved sequences and microRNA function

Four hundred and eighty-one ultraconserved sequences (UCRs) longer than 200 bases were discovered in the genomes of human, mouse and rat. These are DNA sequences showing 100% identity among the three species. UCRs are frequently located at genomic regions involved in cancer, differentially expressed in human leukemias and carcinomas and in some instances regulated by microRNAs (miRNAs). Here we present UCbase & miRfunc, the first database which provides ultraconserved sequences data and shows miRNA function. Also, it links UCRs and miRNAs with the related human disorders and genomic properties. The current release contains over 2000 sequences from three species (human, mouse and rat). As a web application, UCbase & miRfunc is platform independent and it is accessible at http://microrna.osu.edu/.UCbase4.

miR-205 in Breast Cancer: State of the Art

Summary: Despite controversial roles in different cancer types, miR-205 is mainly described as an oncosuppressive miRNA in breast cancer. Its expression decreases from less aggressive to more aggressive subtypes and influences metastasis capability, therapy response, and patient survival. MiR-205 plays a role in breast cancer initiation and progression, stemness maintenance, tumor microenvironment, and may serve as a biomarker with relevance in normal breast physiology. Additionally, some lncRNAs are implicated in breast cancer progression by acting as sponges of miR-205.

Summary: Chronic lymphocytic leukemia (CLL) is a common adult leukemia characterized by chromosomal aberrations. This study found that some CLL patients have undetected microdeletions, which are closely associated with chromosomal alterations in cancer. These findings may have clinical relevance for successful stratification of patients.

Disparities in Lung Cancer: miRNA Isoform Characterization in Lung Adenocarcinoma

Summary: Post-transcriptional modifications in miRNA molecules have provided new insights into cancer biology and potential cancer biomarkers. In this study, we analyzed the profiles of canonical and non-canonical miRNAs in lung cancer patients of different races. We identified distinct post-transcriptional modifications in lung cancer tissues, indicating the relevance of miRNA isoforms as potential biomarkers. Concurrent profiling of canonical and non-canonical miRNAs may help identify additional biological pathways and biomarkers in lung cancer.

Droplet Digital PCR for Non-Invasive Prenatal Detection of Fetal Single-Gene Point Mutations in Maternal Plasma

Summary: In this study, we developed two non-invasive prenatal testing (NIPT) assays for detecting the two most common beta thalassemia mutations in the Mediterranean area. The assays were based on sensitive droplet digital PCR (ddPCR) technology, and were optimized to accurately determine the fetal genotype from maternal plasma samples at different gestational ages. The diagnostic outcomes were confirmed by DNA sequencing. This study provides a simple and sensitive diagnostic tool for NIPT of beta thalassemia mutations, and may have applicability for other single point mutations causing monogenic diseases.

Small Non-Coding RNAs in Soft-Tissue Sarcomas: State of the Art and Future Directions

Summary: Soft-tissue sarcomas (STS), a rare and diverse group of tumors, have various subtypes characterized by differences in tumor biology and clinical outcomes. Small non-coding RNAs (sncRNAs), including miRNAs and tRNA-derived ncRNAs, play important roles in STS tumorigenesis and differentiation. However, the translation of these findings into clinical applications, such as biomarkers or therapeutic targets, remains challenging. This review summarizes the current understanding of sncRNAs in STS, with a focus on liposarcoma, and discusses their potential as novel biomarkers and therapeutic targets.
