Background and Purpose Hypoperfusion-induced thrombosis is an important mechanism for postsurgery stroke and cognitive decline, but there are no perioperative neuroprotectants to date. This study investigated whether prophylactic application of Edaravone, a free radical scavenger already used in treating ischemic stroke in Japan, can prevent infarct and cognitive deficits in a murine model of transient cerebral hypoxia-ischemia. Methods Adult male C57BL/6 mice were subjected to transient hypoxic-ischemic (tHI) insult that consists of 30-minute occlusion of the unilateral common carotid artery and exposure to 7.5% oxygen. Edaravone or saline was prophylactically applied to compare their effects on cortical oxygen saturation, blood flow, coagulation, oxidative stress, metabolites, and learning-memory using methods that include photoacoustic imaging, laser speckle contrast imaging, solid-state NMR, and Morris water maze. The effects on infarct size by Edaravone application at different time points after tHI were also compared. Results Prophylactic administration of Edaravone (4.5 mg/kgx2, IP, 1 hour before and 1 hour after tHI) improved vascular reperfusion, oxygen saturation, and the maintenance of brain metabolites, reducing oxidative stress, thrombosis, white-matter injury, and learning impairment after tHI insult. Delayed Edaravone treatment after 3 h post-tHI became unable to reduce infarct size. Conclusions Acute application of Edaravone may be a useful strategy to prevent postsurgery stroke and cognitive impairment, especially in patients with severe carotid stenosis.

Summary: The study elucidated the role of Poldip2 in vascular inflammation and leukocyte recruitment following cerebral ischemia, showing that Poldip2 depletion attenuated the infiltration of inflammatory cells and decreased the induction of adhesion molecules. Mechanistically, it was demonstrated that FAK activation was a critical intermediary in Poldip2-mediated VCAM-1 induction.

A novel naphthalimide derivative reduces platelet activation and thrombus formation via suppressing GPVI

Summary: The naphthalimide derivative compound 5 exhibits antiplatelet and antithrombotic effects, preventing thrombus formation while potentially increasing bleeding risk. Development of more potent and efficient antiplatelet agents from compound 5 while minimizing bleeding risk may offer therapeutic potential for cardiovascular diseases.

Summary: This study demonstrates the potential of multi-parametric photoacoustic microscopy in quantifying hemodynamics and oxygen metabolism in the mouse brain, allowing longitudinal, cortex-wide, awake-brain imaging. The use of a long-lifetime, wide-field, light-weight, dual-transparency cranial window enabled observation of cerebrovascular responses, which showed complete recovery in 18 days post-installation. Monitoring over 22 weeks post-recovery revealed no adverse changes, indicating the technique's promise for studying chronic progression and therapeutic responses of neurovascular diseases.

Summary: Microglia play a role in regulating vascular tone through PANX1-P2RY12 signaling, with interactions between microglia and brain capillaries crucial for neurovascular structure and function. The study highlights the importance of purines released through PANX1 channels in activating microglial P2RY12 receptors, leading to the regulation of vasodilation and blood flow in the brain.

Summary: Deficiency in the creatine transporter (CrT) can lead to synaptic dysgenesis and signaling imbalances in the brain, particularly under stress conditions. Intranasal creatine supplementation may be an effective treatment for congenital CrT deficiency and acute brain injuries, showing potential in preserving brain energetics homeostasis and reducing infarct size.

Summary: The study demonstrates that adropin can protect the nervous system by maintaining the integrity of the blood-brain barrier, thereby improving brain damage caused by cerebral ischemia and enhancing sensorimotor and cognitive functions. In mouse experiments, adropin treatment or overexpression significantly reduces brain injury, while genetic deletion exacerbates it.
