Summary: In this study, single-cell profiling of human fetal cerebella was performed to investigate the cellular states and regulatory networks in medulloblastomas (MBs). The researchers found a unique transitional cerebellar progenitor population enriched in aggressive MB subgroups and identified transcriptional determinants HNRNPH1 and SOX11 that are correlated with prognosis in group 3 MBs. Genomic analysis revealed long-range chromatin loops connecting HNRNPH1/SOX11-targeted super-enhancers to regulatory elements of the oncogenic driver MYC in group 3 MBs. Targeting the transitional progenitor regulators inhibited MYC expression and MB growth. This study provides new insights into the cellular and molecular characteristics of MBs, which may have implications for disease prognosis and potential therapeutic strategies.

CircRNA circFOXK2 facilitates oncogenesis in breast cancer via IGF2BP3/miR-370 axis

Summary: This study identified Synaptotagmin 11 (SYT11) as a potential therapeutic target and prognostic biomarker in gastric cancer (GC), particularly in the stem-like molecular subtype. SYT11 was found to promote epithelial-mesenchymal transition (EMT) and affect tumor proliferation and metastasis by activating the JNK signaling pathway. Additionally, SYT11 was identified as a potential prognostic biomarker for diffuse-type GC. These findings suggest the potential value of SYT11 as a therapeutic target and prognostic indicator in GC.

Nanofluidics at the crossroads

Summary: Nanofluidics, the study of flows at the smallest scales, has developed rapidly, allowing for fine control of fluidic and ionic transport at the molecular level. Artificial ionic machines can be created by harnessing the phenomena at the nanoscales and taking advantage of techniques from various fields of physics. These machines, based on solid-state nanopores, should exploit their specific properties, such as electronic structure and interaction with light. New methods for probing nanofluidic systems are needed to advance the field and build complex ionic machines inspired by nature.

Summary: MetS is a complex entity that increases the risk of various diseases, including type two diabetes, cardiovascular diseases, and certain types of cancer. miRNAs play an important role in cancer genesis, progression, and metastasis, and the altered secretion of miRs in adipose tissue may explain their involvement in oncogenesis. The interaction between miRs expressed in adipose tissue, their dysregulation, and cancer pathogenesis are still intriguing, as miRNAs exhibit both carcinogenic and tumor suppressor effects.

Childhood B-Cell Preleukemia Mouse Modeling

Summary: Leukemia is the most common cancer in children, with B-cell acute lymphoblastic leukemia (B-ALL) being the most prevalent form. The development of pediatric leukemia is believed to occur through a multi-step or multi-hit mechanism, including prenatal and postnatal steps. Although many initiating events for childhood leukemia occur in utero at a higher frequency than the actual incidence of the disease, the reason why only a small percentage of children with these preleukemic hits develop full-blown leukemia remains unknown. Mouse models that replicate the multi-step process of childhood B-ALL will be crucial in identifying environmental or other factors associated with an increased risk of the disease.

Summary: B cell acute lymphoblastic leukaemia (B-ALL) is the most common form of childhood cancer, but treatment still fails in a significant percentage of patients. Studies have shown that a small percentage of healthy newborns carry preleukaemic clones, but only a small proportion of them will progress to overt leukemia.
