Autism spectrum disorders (ASD) involve a complex interplay of both genetic and environmental risk factors, such as prenatal exposure to valproic acid (VPA). Considering the neuroprotective, antioxidant and anti-inflammatory effects of resveratrol (RSV), we investigated the influence of prenatal RSV treatment on social behaviors of a rodent model of autism induced by prenatal exposure to VPA. In the three-chambered apparatus test, the VPA group showed a reduced place preference conditioned by conspecific and no preference between exploring a wire-cage or a rat enclosed inside a wire cage, revealing sociability impairments. Prenatal administration of RSV prevented the VPA-induced social impairments evaluated in this study. A bioinformatics analysis was used to discard possible molecular interactions between VPA and RSV during administration. The interaction energy between RSV and VPA is weak and highly unstable, suggesting cellular effects instead of a single chemical process. In summary, the present study highlights a promising experimental strategy to evaluate new molecular targets possibly involved in the etiology of autism and developmental alterations implicated in neural and behavioral impairments in ASD. Binding site hotspot map of PI3Kα and mTOR in the presence of selective and dual ATP-competitive inhibitors

Francisca Fernanda Nunes Azevedo, Francisca Joseli Freitas de Sousa, Francisco Lucas Santos de Oliveira, Jaqueline Vieira Carletti, Geancarlo Zanatta

Summary: The study compared the ATP-binding pockets of PI3K alpha and mTOR using existing crystallographic structures and investigated their binding profiles and relative contributions in the presence of selective or dual inhibitors. The results showed that the most important interactions occurred within a range of 3-3.5 angstroms, with residues within this range contributing to 75-100% of the total calculated interaction energy. The study provides key information for understanding the molecular mechanisms of multi-target inhibition.

Summary: Atrazine, a widely used herbicide, has been found to cause persistent contamination of water and soil due to its high resistance to degradation. It is associated with fertility issues, increased risk of prostate cancer, and birth defects. This study characterizes the binding of atrazine to human serum albumin (HSA) and identifies the most likely binding sites and the amino acid residues involved in the binding interactions.

Summary: This study investigates the structural, electronic, and optical properties of DMF using density functional theory (DFT). The results show that the delta-sol correction method improves the estimation of the band-gap energy from DFT calculations. Additionally, simulations of a monolayer and crystal form of DMF demonstrate that intermolecular interactions decrease the electronic energy gap.

GABAergic synaptic transmission and cortical oscillation patterns in the primary somatosensory area of a valproic acid rat model of autism spectrum disorder

Summary: Prenatal exposure to valproic acid (VPA) leads to autism spectrum disorder (ASD), but resveratrol (RSV) can prevent these impairments. This study found that prenatal VPA exposure reduced synaptic inhibition in the somatosensory area, resulting in behavioral and electroencephalogram changes. However, RSV prevented these synaptic inhibition alterations and improved behavioral and electroencephalogram abnormalities.

The lipase from Thermomyces lanuginosus was found to exhibit high enantioselectivity towards the acetylation of (Sp)-1, leading to the preferential formation of (Sp)-2. Both (Rp)-1 and (Sp)-2 showed low cytotoxicity and no selectivity against the tested cell lines.

Neural correlates of social interaction anxiety and their relation to emotional intelligence: A resting-state fMRI study

Song Xue, Feng Kong, Yiying Song, Jia Liu

Summary: This study used resting-state functional magnetic resonance imaging to explore the relationship between individual's spontaneous neural activity and social interaction anxiety in a nonclinical population. The results showed that social interaction anxiety was correlated with the fractional amplitude of low-frequency fluctuations in several brain regions, and that emotional intelligence partially mediated this relationship. This study provides evidence for the neural basis of social interaction anxiety in the normal population and highlights the role of emotional intelligence in this anxiety.

Development of the human medullary arcuate nucleus from mid-gestation to the perinatal period: A morphometric study

Katsuyuki Yamaguchi, Takuya Yazawa

Summary: This study provides morphometric data on the development of the human medullary arcuate nucleus (AN) by examining the brains of preterm and perinatal infants. The results show that AN morphology demonstrates asymmetry and individual variability during the fetal period. The volume and neuronal number of AN increase exponentially with age, while neuronal density decreases exponentially. The AN may undergo neuron death and neuroblasts production after mid-gestation.

Alamandine injection in the periaqueductal gray and rostral ventromedial medulla attenuates allodynia induced by sciatic nerve ligation in rats

Summary: This study investigated the effects of alamandine on allodynia in a rat model and found the presence of MrgD receptors in the vlPAG and RVM regions. Microinjection of alamandine resulted in a significant increase in paw withdrawal threshold and could be blocked by an MrgD receptor antagonist. Upregulation of MrgD receptor expression following allodynia induction suggests a potential compensatory mechanism in response to pain.

Evaluation of the separate and combined effects of anodal tDCS over the M1 and F3 regions on pain relief in patients with type-2 diabetes suffering from neuropathic pain

Summary: The present study aimed to investigate the separate and combined effects of anodal transcranial direct current stimulation (tDCS) over the primary motor cortex (M1) and left dorsolateral prefrontal cortex (F3) regions on pain relief in patients with type-2 diabetes suffering from neuropathic pain (NP). The results showed that tDCS had the potential to induce pain relief in patients with type-2 diabetes suffering from NP. The mean perceived pain intensity in the posttest was lower in the M1 stimulation group than in the F3 stimulation group. However, more trials with larger sample sizes are necessary to define clinically relevant effects.

The cognitive architecture of verbal humor in schizophrenia

Summary: This study found a general impairment in humor comprehension in individuals with schizophrenia, with mental jokes being more difficult for both patients and controls. Humor comprehension was closely associated with the patients' overall pragmatic and linguistic profile, while the association with Theory of Mind (ToM) was minimal. Another notable finding was the increased appreciation of humor in individuals with schizophrenia, who rated jokes as funnier than controls did, regardless of whether they were correctly or incorrectly completed. The funniness ratings were not predicted by any measure, suggesting a dimension of humor untied to cognition or psychopathology.
