The present study was performed to explore the effect of green tea polyphenols on the intracellular A beta (iA beta)-induced toxicity to cultured rat primary prefrontal cortical neurons. Administration of 100 nM, 1 mu M or 10 mu M of green tea polyphenols significantly inhibited the iA beta-induced toxicity to cultured rat primary prefrontal cortical neurons tested by MTT and LDH release assays. We further studied the involvement of neuroprotective pathway protein AKT in green tea polyphenols protection against iA beta-induced cytotoxicity on cultured rat primary prefrontal cortical neurons. The results demonstrated that the content of p-AKT decreased significantly after iA beta treatment, while administration of green tea polyphenols significantly inhibited the iA beta-induced decrease in the content of p-AKT. Moreover, blockade of AKT signalling inhibited the protective effects of green tea polyphenols against iA beta-induced neurotoxicity. The results suggest that green tea polyphenols may play a protective effect on cultured rat primary prefrontal cortical neurons against iA beta-induced cytotoxicity and AKT is involved in the green tea polyphenols-induced protective effects. Hua Liu, Shan Lan, Xiao-Jie Shi, Fang-Cheng Fan, Qing-Shan Liu, Lin Cong, Yong Cheng

Summary: MicroRNAs (miRNAs) play a crucial role in the development of neurodegenerative disorders, particularly amyotrophic lateral sclerosis (ALS). Through a comprehensive systematic review and meta-analysis, the study identified elevated levels of miR-206, miR-133b, and miR-338-3p in patients with ALS compared to healthy controls. These findings suggest that miR-206, miR-133b, and miR-338-3p may serve as potential promising biomarkers for ALS, indicating their importance in the pathogenesis and progression of the disease.

Risk Factors for Postpartum Depression Based on Genetic and Epigenetic Interactions

Fan Luo, Zimo Zhu, Yang Du, Lei Chen, Yong Cheng

Summary: Postpartum depression is a serious mood disorder that occurs after childbirth and can have lifelong consequences for women and their families. It is influenced by various risk factors, including environmental and genetic factors. Gene-environment interactions play a role in postpartum depression, as evidenced by studies on genes related to neurotransmitters, the HPA axis, and the kynurenine pathway. However, the conclusions regarding these risk factors, especially genetic factors, are not consistent, and further research is needed to understand the heritability and susceptibility of postpartum depression.

Neurometabolite levels in the brains of patients with autism spectrum disorders: A meta-analysis of proton magnetic resonance spectroscopy studies (N=1501)

Yang Du, Lei Chen, Mei-Chen Yan, Yan-Li Wang, Xiao-Lin Zhong, Chen-Xi Xv, Yao-Bo Li, Yong Cheng

Summary: A meta-analysis of 54 studies revealed that patients with autism spectrum disorders (ASDs) have altered levels of neurometabolites in specific brain regions compared to healthy controls. Lower levels of N-acetyl-aspartate-containing compound (NAA), choline-containing compound (Cho), and NAA/(creatine-containing compound) Cr ratios in the gray matter, as well as lower levels of NAA and glutamate + glutamine (Glx) in the white matter, were observed in ASD patients. Additionally, the frontal cortex showed decreased NAA and gamma-aminobutyric acid (GABA) levels, NAA/Cr ratios, and increased glutamate (Glu) levels. These findings suggest that neurometabolite alterations in specific brain regions are associated with ASD, and age is a confounding factor for certain neurometabolite levels in patients with ASD.

MicroRNA schizophrenia: Etiology, biomarkers and therapeutic targets

Heng-Chang Zhang, Yang Du, Lei Chen, Zeng-Qiang Yuan, Yong Cheng

Summary: Schizophrenia-related symptoms, including positive, negative, and cognitive symptoms, have serious effects on public health, affecting approximately 1% of the population. Recent studies have shown that besides the traditional dopaminergic pathway, the etiology of schizophrenia involves neuronal networks such as glutamate, GABA, serotonin, BDNF, oxidative stress, inflammation, and the immune system. MicroRNAs (miRNAs), small noncoding RNA molecules, have been identified as potential factors in the pathophysiology of schizophrenia, influencing these systems. Understanding how changes in miRNAs affect critical signaling pathways could lead to improved approaches in treating and diagnosing schizophrenia.

Summary: This study found a general impairment in humor comprehension in individuals with schizophrenia, with mental jokes being more difficult for both patients and controls. Humor comprehension was closely associated with the patients' overall pragmatic and linguistic profile, while the association with Theory of Mind (ToM) was minimal. Another notable finding was the increased appreciation of humor in individuals with schizophrenia, who rated jokes as funnier than controls did, regardless of whether they were correctly or incorrectly completed. The funniness ratings were not predicted by any measure, suggesting a dimension of humor untied to cognition or psychopathology.
