Scaffolding proteins in highly purified rat olfactory cilia membranes

Odour-mediated signal transduction is a complex process that occurs in the cilia of olfactory sensory neurons. To gain insight in to the molecular organization of the odour transduction machinery, we developed a procedure to purify olfactory cilia membranes by differential centrifugation of rat olfactory epithelium extracts. We tested whether known scaffolding proteins that might participate in the assembly of the complex chemotransduction apparatus are present in the purified membrane fraction. Utilizing immunoblotting and immunohistochemistry, we show that the multidomain scaffolding proteins ProSAP/Shanks and calcium/ calmodulin-dependent serine protein kinase CASK are present in the olfactory cilia. Ion channels involved in chemotransduction could be reconstituted into planar lipid bilayers for electrophysiological recordings. Summary: The study demonstrates the possibility of producing PHB as a fermentation product in E. coli with higher efficiency. The importance of an acetoacetyl-CoA reductase with a preference for NADH in PHB production is highlighted through structural and kinetic analyses. The assembly of an operon combining phaCA genes with the gene encoding the NADH-preferring acetoacetyl-CoA reductase results in continuous accumulation of PHB in E. coli cells under oxygen limiting conditions, indicating opportunities for further protein/metabolic engineering strategies.

The results revealed 287 distinct genomic loci and 120 genes likely to be involved in the development of schizophrenia. Further research in this area could provide insights into new therapeutic approaches for skeletal disorders.

Summary: TRPV4 is associated with various diseases, and mitochondrial abnormalities play a significant role in these diseases. Improper expression and function of TRPV4 lead to mitochondrial abnormalities, and TRPV4 also regulates mitochondrial numbers, Ca2+ levels, and temperature. Naturally occurring TRPV4 mutations affect mitochondrial morphology and distribution, which helps in understanding TRPV4-mediated channelopathies and classifying them as mitochondrial diseases.

Summary: Synapse diversity has been described from different perspectives, ranging from the specific neurotransmitters released, to their diverse biophysical properties and proteome profiles. However, synapse diversity at the transcriptional level has not been systematically identified across all synapse populations in the brain. To quantify and identify specific synaptic features of neuronal cell types, the SynGO database was combined with single-cell RNA sequencing data of the mouse neocortex. This study provides a deeper understanding of synapse diversity in the neocortex and identifies potential markers to selectively identify synapses from specific neuronal populations.

Design and thermodynamic analysis of a pathway enabling anaerobic production of poly-3-hydroxybutyrate in Escherichia coli

Summary: This study proposes a novel metabolic engineering strategy to achieve efficient anaerobic production of poly-(R)-3-hydroxybutyrate (PHB) in Escherichia coli. The strategy involves rerouting the glycolytic flux through non-oxidative glycolysis and coupling PHB synthesis with NADH re-oxidation. Stoichiometric analysis and various thermodynamic approaches were used to assess the feasibility and bottlenecks of the engineered pathway.
