Summary: Co-pathologies are important in the expression of the clinical phenotype of Alzheimer's disease, with differences in neuropathological diagnoses between early-onset and late-onset patients. Late-onset Alzheimer's disease is more likely to have LATE, hippocampal sclerosis, argyrophilic grain disease, and vascular brain injury.Sex does not significantly impact the number of co-pathologies present.

Secretory autophagy maintains proteostasis upon lysosome inhibition

Tina A. Solvik, Tan A. Nguyen, Yu-Hsiu Tony Lin, Timothy Marsh, Eric J. Huang, Arun P. Wiita, Jayanta Debnath, Andrew M. Leidal

Summary: The endolysosome system is involved in autophagic degradation and secretory pathways, including the release of extracellular vesicles and particles (EVPs). This study reveals a secretory autophagy pathway that is upregulated during endolysosome inhibition and mediates the release of autophagic cargo receptors via EVPs. This secretion process is regulated by multiple ATGs and the small GTPase Rab27a.

Comment on Impact of neurodegenerative diseases on human adult hippocampal neurogenesis

Arturo Alvarez-Buylla, Arantxa Cebrian-Silla, Shawn F. Sorrells, Marcos Assis Nascimento, Mercedes F. Paredes, Jose Manuel Garcia-Verdugo, Zhengang Yang, Eric J. Huang

Summary: This study lacks evidence to support the link between proliferating cells and neurogenesis, the role of multipolar nestin(+) astrocytes as progenitors, and the identity of mature-looking doublecortin+ neurons as adult-born. The histology-marker expression observed differs from species with well-documented adult hippocampal neurogenesis.

Summary: BRAT1 encephalopathy is an ultra-rare autosomal recessive neonatal encephalopathy characterized by multifocal myoclonus, microcephaly, hypertonia, and multifocal seizures. Early recognition of BRAT1 encephalopathy is crucial for prompt workup, appropriate management, and genetic counseling.

Disentangling brain vasculature in neurogenesis and neurodegeneration using single-cell transcriptomics

Elizabeth E. Crouch, Tara Joseph, Elise Marsan, Eric J. Huang

Summary: This article discusses the impact of vasculature on brain function, including angiogenesis and neurogenesis during embryonic brain development and neurovascular interactions in the adult brain to maintain brain function. The article also explores the use of single-cell transcriptomics to study the subtypes, organization, and zonation of vascular cells in the embryonic and adult brain, as well as the role of dysfunction in neurovascular and gliovascular interactions in the pathogenesis of neurodegenerative diseases. Finally, the article highlights key challenges for future research in neurovascular biology.

Profiling human brain vascular cells using single-cell transcriptomics and organoids

Elizabeth E. Crouch, Loukas N. Diafos, Edward J. Valenzuela, Kaylee Wedderburn-Pugh, Janeth Ochoa Birrueta, Jaela Caston, Tara Joseph, Madeline G. Andrews, Aparna Bhaduri, Eric J. Huang

Summary: Angiogenesis and neurogenesis are functionally interconnected during brain development. This article presents a method to purify prenatal human brain vascular cells using FACS and utilize them in various downstream applications. The method is simple and efficient, generating high yields from small amounts of tissue. These vascular cells can be cultured and used in various in vitro assays. Additionally, the article describes how to use these cells for transplantation into cortical organoids to study neurovascular interactions in prenatal human brain development.
