Gentile, E. Nobile-Orazio

Summary: The study found that in patients with clinical CIDP diagnosis but not meeting EFNS/PNS criteria, multiple abnormal supportive criteria helped to support the diagnosis of CIDP. Increased cerebrospinal fluid proteins and response to immune therapy were the main factors aiding in the diagnosis of CIDP.

A diagnostic score for anti-myelin-associated-glycoprotein neuropathy or chronic inflammatory demyelinating polyradiculoneuropathy in patients with anti-myelin- associated-glycoprotein antibody

Pietro E. Doneddu, Marta Ruiz, Elisa Bianchi, Giuseppe Liberatore, Fiore Manganelli, Dario Cocito, Giuseppe Cosentino, Luana Benedetti, Girola A. Marfia, Massimiliano Filosto, Chiara Briani, Claudia Giannotta, Eduardo Nobile-Orazio

Summary: A diagnostic score was developed to distinguish anti-MAG neuropathy from CIDP and was applied to patients with atypical anti-MAG neuropathy. The score allowed an accurate discrimination between the two groups and may help in selecting proper treatment for patients with anti-MAG antibodies.

Loss of thymidine phosphorylase activity disrupts adipocyte differentiation and induces insulin-resistant lipoatrophic diabetes

Jeremie Gautheron, Lara Lima, Baris Akinci, Jamila Zammouri, Martine Auclair, Sema Kalkan Ucar, Samim Ozen, Canan Altay, Bridget E. Bax, Ivan Nemazanyy, Veronique Lenoir, Carina Prip-Buus, Cecile Acquaviva-Bourdain, Olivier Lascols, Bruno Feve, Corinne Vigouroux, Esther Noel, Isabelle Jeru

Summary: This study identifies the impact of TYMP variants on Mitochondrial NeuroGastroIntestinal Encephalomyopathy (MNGIE) and lipodystrophic diabetes. Using CRISPR-Cas9, researchers demonstrate that TP knockout leads to impaired adipocyte differentiation, decreased insulin response, oxidative stress, altered mitochondrial functions, and cellular senescence. These findings highlight the crucial regulatory role of TP in adipose tissue biology.

Summary: This study compared the sensitivity and specificity of two diagnostic criteria (EAN/PNS and EFNS/PNS) for chronic inflammatory demyelinating polyradiculoneuropathy (CIDP). The results showed that the EAN/PNS criteria had higher specificity but lower sensitivity compared to the EFNS/PNS criteria, with the latter being more sensitive for the diagnosis of typical CIDP.

Summary: This study assessed the ability of the 2021 European Academy of Neurology/Peripheral Nerve Society (EAN/PNS) clinical criteria for chronic inflammatory demyelinating polyradiculoneuropathy (CIDP) to classify the whole spectrum of clinical heterogeneity of the disease and define the clinical characteristics of unclassifiable clinical forms. The results showed that a proportion of patients with CIDP did not strictly fulfill the 2021 EAN/PNS clinical criteria, and these unclassifiable clinical phenotypes may pose diagnostic challenges and deserve more attention in clinical practice and research.

Effects of Triheptanoin on Mitochondrial Respiration and Glycolysis in Cultured Fibroblasts from Neutral Lipid Storage Disease Type M (NLSD-M) Patients

Nelida Ines Noguera, Daniela Tavian, Corrado Angelini, Francesca Cortese, Massimiliano Filosto, Matteo Garibaldi, Sara Missaglia, Ariela Smigliani, Alessandra Zaza, Elena Maria Pennisi

Summary: Neutral lipid storage disease type M (NLSD-M) is an ultra-rare, autosomal recessive disorder that causes severe skeletal and cardiac muscle damage and lipid accumulation in all body tissues. Triheptanoin has been shown to induce an anaplerotic effect in NLSD-M fibroblasts, indicating potential for new therapeutic strategies.

A Coxsackievirus B1-mediated nonlytic Extracellular Vesicle- to-cell mechanism of virus transmission and its possible control through modulation of EV release

Samireh Jorfi, Ephraim Abrokwa Ansa-Addo, Katia Mariniello, Purva Warde, Ahmad Asyraf bin Senian, Dan Stratton, Bridget E. Bax, Michelle Levene, Sigrun Lange, Jameel Malhador Inal

Summary: CVB1, a non-enveloped virus, spreads using cell lysis and extracellular vesicles (EVs). In this study, it was discovered that CVB1 induces apoptosis and releases CVB1-induced 'medium-sized' EVs (CVB1i-mEVs) that contain the virus. These EVs disseminate CVB1 to new host cells through a non-lytic EV-to-cell mechanism.

Prognostic Usefulness of Motor Unit Number Index (MUNIX) in Patients Newly Diagnosed with Amyotrophic Lateral Sclerosis

Barbara Risi, Stefano Cotti Piccinelli, Stefano Gazzina, Beatrice Labella, Filomena Caria, Simona Damioli, Loris Poli, Alessandro Padovani, Massimiliano Filosto

Summary: The MUNIX technique can estimate the number and size of surviving motor units (MUs). Previous studies found correlations between MUNIX and several clinical measures in ALS patients, but its potential as a predictor of disease progression rate (DPR) has not been thoroughly evaluated. Our study aimed to investigate the ability of MUNIX to predict DPR at a six-month follow up. The results showed that MUNIX could predict DPR in ALS patients and could be used as a prognostic tool.

Juvenile-Onset Recurrent Rhabdomyolysis Due to Compound Heterozygote Variants in the ACADVL Gene

Beatrice Labella, Gaetana Lanzi, Stefano Cotti Piccinelli, Filomena Caria, Simona Damioli, Barbara Risi, Enrica Bertella, Loris Poli, Alessandro Padovani, Massimiliano Filosto

Summary: This article discusses a rare genetic disorder called Very long-chain acyl-CoA dehydrogenase (VLCAD) deficiency, caused by mutations in the ACADVL gene. A case study of a 17-year-old boy with exercise-induced muscle pain and fatigue, as well as other symptoms, is reported. Various tests confirmed the diagnosis of VLCAD deficiency, and early dietary treatment is crucial for minimizing muscle damage and slowing disease progression.

Summary: Pompe disease is a genetic disorder caused by mutations in the GAA gene that result in glycogen storage in lysosomes, particularly in muscle tissue. The relationship between genotype and phenotype has been extensively discussed, suggesting that other genetic variants may influence the clinical presentation. In this study, we identified potentially relevant variants in genes related to glycogen synthesis and catabolism in late-onset Pompe disease patients.
