Effects of the COVID-19 pandemic on acute coronary syndromes in Germany during the first wave: the COVID-19 collateral damage study

Summary: This study aimed to comprehensively describe hospital admissions and in-hospital mortality during the first wave of the COVID-19 pandemic in all German federal states. The results showed a significant reduction in admissions for NSTEMI and STEMI, while the quality of in-hospital care and in-hospital mortality were not affected.

Summary: Protein translation is crucial for cell function, and disruptions in this process can lead to neurodevelopmental disorders. The eEF1A and eEF1B complex are involved in delivering aminoacyl-tRNA to the ribosome. Mutations in the gene EEF1D, which encodes a subunit of the eEF1B complex, have been linked to severe intellectual disability and microcephaly. We identified new variants in EEF1D in two families with developmental delay, microcephaly, spasticity, and other symptoms. These variants affect the GEF domain of EEF1D, suggesting a potential link between disturbed protein translation machinery and neurodevelopmental phenotypes.

Sex-specific Inequalities in the Treatment of Severely Calcified Coronary Lesions: A Sub-analysis of the PREPARE-CALC Trial

Summary: This study investigated the sex-specific characteristics and outcomes of patients with calcified coronary arteries in the Prepare-CALC trial. The results showed that rotational atherectomy was superior to modified balloon therapy in men, while both strategies had similar success rates in women. There were no significant differences in complications between sexes or treatment strategies.

Homozygous loss-of-function variants in FILIP1 cause autosomal recessive arthrogryposis multiplex congenita with microcephaly

Summary: This study reports five patients from three families with a disorder characterized by congenital joint contractures affecting multiple body parts, including shoulder, elbow, hand, hip, knee, and foot, as well as scoliosis, reduced palmar and plantar skin folds, microcephaly, and facial dysmorphism. Exome sequencing identified homozygous truncating variants in the FILIP1 gene in all patients. FILIP1 is involved in the regulation of filamin homeostasis and is essential for cortical cell migration and muscle cell differentiation.

Summary: By integrating large-scale whole-genome sequencing data, transcriptomic data, and epigenomic data, researchers found that the current World Health Organization classification only explains 10% of the molecular differences between patients. Instead, the MESOMICS project establishes a morphomolecular classification of malignant pleural mesothelioma based on four dimensions: ploidy, tumor cell morphology, adaptive immune response, and CpG island methylator profile. These findings reveal the interplay between MPM functional biology and its genomic history and provide new insights into the variations observed in the clinical behavior of MPM patients.

The genetic spectrum of congenital ocular motor apraxia type Cogan: an observational study, continued

Summary: Congenital ocular motor apraxia (COMA) is a heterogeneous eye movement disorder with various underlying genetic causes. In this study, researchers re-evaluated the neuroimaging features of 21 patients with COMA and discovered a novel sign, molar tooth sign (MTS), leading to a reassignment of diagnosis to Joubert syndrome (JBTS) in some patients. Using genetic analysis, pathogenic variants were identified in 81% of the patients, involving nine different genes. A diagnostic algorithm for COMA was proposed based on these findings.

Summary: This study identified a novel molecular chaperonopathy causing impaired protein O-glycosylation. The patients had decreased activity of T-synthase, an enzyme that synthesizes the T-antigen. The chaperone Cosmc, encoded by the C1GALT1C1 gene, was found to be responsible for the T-synthase function.

Summary: Despite improvements in therapies, the outcomes for patients with acute myocardial infarction (AMI) and cardiogenic shock (CS) remain poor. CS occurs in 3-15% of AMI cases, with high mortality rates. Limited evidence-based knowledge and nonuniform management lead to underutilization or overutilization of treatment options. This article aims to provide a critical overview of the literature on CS management, summarizing recent evidence on revascularization strategies, pharmacological treatments, and mechanical circulatory support (MCS) use.

Summary: This study investigated the prognostic value and clinical utility of left atrial reservoir strain (LARS) in patients undergoing transcatheter aortic valve replacement (TAVR) for aortic stenosis (AS). The results showed that impaired LARS was independently associated with worse outcomes, and LARS assessment allowed for reliable prediction in patients where conventional echocardiographic assessment was unavailable.
