Transient Receptor Potential Vanilloid 4 blockade protects against experimental colitis in mice: a new strategy for inflammatory bowel diseases treatment?

Recent reports suggested that the activation of Transient Receptor Potential Vanilloid 4 (TRPV4) receptors in the gastrointestinal tract has pro-inflammatory effects. In this study, we demonstrated for the first time that TRPV4 mRNA expression is up-regulated in patients with inflammatory bowel diseases (IBD). Furthermore, selective blockade of TRPV4 in the 2,4,6-trinitrobenzenesulfonic acid animal model alleviates colitis and pain associated with the intestinal inflammation. Our study indicates that TRPV4 may play a role in mechanisms of defense in intestinal inflammation and that TRPV4 may be an attractive target for future systemic or topic anti-inflammatory treatment in patients with IBD.

COMPLEX-FORMING PROPERTIES OF SYNTHETIC PYRIMIDINONE NUCLEOSIDES WITH Ag(I) IONS IN AN AQUEOUS SOLUTION

Summary: The complexation properties of two synthetic nucleosides, beta-D-ribofuranoside-4-pyrimidinone (ribo) and beta-D-deoxyribofuranoside-4-pyrimidinone (deoxy), with Ag(I) ions in aqueous solution were investigated. The stability constants of the coordination entities formed were calculated and the percentage of each entity in solution was estimated. The results suggest that these coordination entities have potential antiviral, antibacterial, and antitumor properties, as well as potential applications in nanobiotechnology.

A New Gold(III) Complex, TGS 703, Shows Potent Anti-Inflammatory Activity in Colitis via the Enzymatic and Non-Enzymatic Antioxidant System-An In Vitro, In Silico, and In Vivo Study

Summary: Inflammatory bowel diseases (IBD), such as Crohn's disease and ulcerative colitis, are global health problems with increasing frequency and unclear pathogenesis. Current treatment involves drugs like corticosteroids and derivative compounds, but more specific and effective therapies are desired. In this study, novel gold complexes were tested and found to have anti-inflammatory potential both in vitro and in vivo, particularly TGS 703, which was able to alleviate inflammation in a mouse model of colitis and showed a link to antioxidant systems. These findings suggest that gold(III) complexes may have therapeutic applications in the treatment of IBD.

Putative molecular targets for vitamin A in neutralizing oxidative stress in acute and chronic pancreatitis - a systematic review

Summary: Acute pancreatitis (AP) and chronic pancreatitis (CP) are severe diseases that pose a great threat to human health in high-income countries. Oxidative stress plays a significant role in the development of AP and CP, and there is growing interest in the use of antioxidant therapy to manage these conditions. Vitamin A, a well-known antioxidant, has been understudied in relation to its impact on oxidative stress in AP and CP. This review aims to evaluate the molecular targets of vitamin A and its potential role in attenuating oxidative stress in AP and CP. The findings suggest that vitamin A may influence several molecular pathways involved in antioxidative defense and cytoprotection, indicating its potential to alter the disease course of AP and CP.

Summary: Metal-based agents in cancer therapy, like cisplatin and its derivates, have established clinical applications but also can induce serious side effects. Thus, metallotherapeutic alternatives for platinum derivatives are developed and intensively studied. Platinum is replaced by several transition metals including gold. Especially gold (III) complexes can have the same square-planar structure and are isoelectric with platinum (II).

Empagliflozin attenuates intestinal inflammation through suppression of nitric oxide synthesis and myeloperoxidase activity in in vitro and in vivo models of colitis

Summary: Inflammatory bowel diseases (IBD) is a chronic and relapsing inflammation in the gastrointestinal tract. The incidence and prevalence of IBD are high and increasing, and current therapeutic strategies are suboptimal. This study focused on gliflozins, a group of anti-diabetic drugs, and found that they have anti-inflammatory properties. In vitro experiments showed that two gliflozins, EMPA and DAPA, suppressed nitric oxide production in macrophages, and in vivo experiments demonstrated that EMPA alleviated colitis in mice. Our findings suggest that SGLT-2 inhibitors have anti-inflammatory effects independent of their hypoglycemic effects.

CA 19-9 but Not IGF-1/IGFBP-2 Is a Useful Biomarker for Pancreatic Ductal Adenocarcinoma (PDAC) and Chronic Pancreatitis (CP) Differentiation

Summary: Differentiating between pancreatic ductal adenocarcinoma (PDAC) and chronic pancreatitis (CP) poses a challenge. This study assessed the usability of IGF-1, IGFBP-2, and IGF-1/IGFBP-2 ratio in differentiating PDAC and CP. The results indicate that CA 19-9 demonstrates high potential as a marker for PDAC and CP differentiation.

Diagnosis and Management of Barrett's Esophagus

Summary: Barrett's esophagus is a precancerous condition of the esophagus, characterized by a salmon-colored lining and the presence of columnar epithelium with goblet cells. It is associated with esophageal adenocarcinoma, a tumor with poor survival rates and increasing incidence. The diagnosis and management of Barrett's esophagus remain a topic of ongoing debate. This article provides an overview of current recommendations and new discoveries in this field.

Eosinophilic Esophagitis-What Do We Know So Far?

Summary: Eosinophilic esophagitis is a Th-2 antigen-mediated disease that causes inflammation in the esophagus with an influx of eosinophils. It leads to esophageal remodeling and difficulties in swallowing. While the disease has a relatively short history, its incidenc e and prevalence are increasing rapidly, especially in Western countries. The diagnosis requires the presence of at least 15 eosinophils per high-power field in esophageal biopsies, along with relevant symptoms and exclusion of other causes of esophageal eosinophilia. Treatment options for eosinophilic esophagitis are challenging, but the combination of proton pump inhibitors, topical corticosteroids, elimination diet, and biological treatment has shown promise. New methods for disease diagnosis and clinical assessment are also available.

Relation between Selected Sleep Parameters, Depression, Anti-Tumor Necrosis Factor Therapy, and the Brain-Derived Neurotrophic Factor Pathway in Inflammatory Bowel Disease

Summary: Inflammatory bowel disease (IBD) patients often experience sleep and mood disorders which may be related to the disruption of the brain-gut axis. This study found that IBD patients had lower expression of BDNF mRNA but higher concentrations of proBDNF and BDNF protein compared to healthy controls. BDNF protein levels were positively correlated with sleep efficiency in the IBD group and depression severity was associated with BDNF mRNA and protein levels in the remission group.

Evaluation of Albumin, Transferrin and Transthyretin in Inflammatory Bowel Disease Patients as Disease Activity and Nutritional Status Biomarkers

Summary: This study evaluated the utility of albumin, transferrin, and transthyretin levels in assessing nutritional status and activity of inflammatory bowel disease (IBD). The results found significantly lower albumin levels in IBD patients compared to controls and in Crohn's disease (CD) patients compared to ulcerative colitis (UC) patients. Higher transthyretin levels were found in IBD patients compared to healthy subjects. Albumin levels in IBD patients were associated with disease activity and nutritional status, while further studies are needed to assess the utility of transferrin and transthyretin as markers.

The Latest Advancements in Diagnostic Role of Endosonography of Pancreatic Lesions

Summary: Endosonography, a minimally invasive imaging technique, has revolutionized the diagnosis and management of pancreatic diseases. This review highlights the latest advancements in endosonography of the pancreas, including key technological developments, procedural techniques, clinical applications and additional techniques. EUS is well established for T-staging and N-staging of pancreaticobiliary malignancies, for pancreatic cyst discovery, for identifying subepithelial lesions (SEL), for differentiation of benign pancreaticobiliary disorders or for acquisition of tissue by EUS-guided fine-needle aspiration or EUS-guided fine-needle biopsy. This review briefly describes principles and application of EUS and its related techniques.

The Effect of Serotonin Transmission on Depressive and Insomnia Symptoms in Inflammatory Bowel Diseases

Summary: The study investigates the impact of the serotonergic pathway on the pathogenesis and course of inflammatory bowel diseases (IBDs), and its relationship with sleep and mood disorders. The results show that patients with active IBD have higher levels of 5-HT and SERT protein expression, which are positively correlated with insomnia severity. After treatment, only 5-HT levels are decreased.

Clinical and Laboratory Characteristics of Anaemia in Hospitalized Patients with Inflammatory Bowel Disease

Summary: This study aimed to evaluate the usefulness of laboratory parameters, including hepcidin, in the differential diagnosis of anaemia in hospitalized patients with inflammatory bowel disease (IBD). The study included 118 adult patients with IBD – 55 with ulcerative colitis (UC) and 63 with Crohn's disease (CD). Anaemia was more prevalent in CD patients compared to UC patients, and its prevalence increased with the severity of IBD and inflammatory changes. Comprehensive laboratory assessment, including hepcidin levels, is necessary for the full picture of anaemia in IBD.
