Summary: A phase 3 multicentre, observer-blind, randomised, controlled trial was conducted to evaluate the efficacy, immunogenicity, and safety of a toxoid vaccine candidate for Clostridioides difficile infection. However, the study was terminated after showing that the bivalent C difficile toxoid vaccine did not prevent C difficile infection, leading to the discontinuation of its clinical development.

Summary: This study describes a new method for purifying, expanding, and retrovirally transducing mouse Tregs, and confirms the feasibility of the method in experiments. By using this method, it is possible to study transduced Tregs in animal models, providing a new avenue for research on genetically engineered Treg therapy for various inflammatory diseases.

Summary: This study aimed to evaluate the safety and efficacy of sarilumab in patients with severe or critical COVID-19, but found no significant efficacy in patients admitted to the hospital receiving supplemental oxygen. Future adequately powered trials of targeted immunomodulatory therapies with survival as a primary endpoint are suggested for patients with critical COVID-19.

An in vitro chronic damage model impairs inflammatory and regenerative responses in human colonoid monolayers

Summary: We developed a chronic-injury model to study the effects of chronic intestinal damage on intestinal epithelial cells (IECs). We found that repeated rounds of damage impair the regrowth of IECs and their response to microbial stimulation. We also observed changes in mRNA expression and DNA methylation in genes associated with inflammatory bowel disease (IBD) and colon cancer.

Summary: Microbial exposures play a crucial role in impacting healthspan through shaping the immune system and microbiota. The use of Phage ImmunoPrecipitation Sequencing (PhIP-Seq) allows for high-throughput and cost-effective detection of exposure and response to microbial protein products. This study demonstrates that PhIP-Seq with the ToxScan library is an effective tool for studying the environmental determinants of health and disease at a cohort scale.

Summary: This study evaluated the real-time polymerase chain reaction (qPCR) cycle threshold (CT) as a predictor for disease severity and TcdB enzyme immunoassay (EIA) results. The results showed that CT value was an acceptable predictor for EIA toxin positivity but had limited predictability for clinical severity.

Summary: GSK3 beta inhibitors have shown broad-spectrum anti-coronaviridae activity, including against SARS-CoV-2, with low toxicity to host cells. A lead compound, T-1686568, exhibited dose-dependent activity against SARS-CoV-2 and HCoV-229E and showed efficacy in viral-infected cells and organoids. GSK3 beta and PKCa can phosphorylate the nucleocapsid protein of SARS-CoV-2, inhibiting its translation.
