Class I Histone Deacetylase Inhibitor Valproic Acid Reverses Cognitive Deficits in a Mouse Model of Septic Encephalopathy

Accumulating evidence suggests that histone deacetylase inhibitor exert neuroprotective effects in animal models of neurological diseases. We investigated for the first time whether class I histone deacetylase inhibitor valproic acid (VPA) can reverse cognitive deficits in a mouse model of sepsis-associated encephalopathy (SAE). Moreover, the possible mechanisms of protection were also explored. A mouse model of SAE was induced in adult male mice by cecal ligation and puncture (CLP). Mice received an administration of saline or VPA (100 mg/kg) once daily for 14 consecutive days starting either immediately or 2 weeks after operation. Furthermore, the TrkB antagonist K252a was used in another group of experiment to investigate whether brain-derived neurotrophic factor (BDNF)-TrkB signaling pathway is involved in the protection of VPA. Our data suggested that CLP resulted in significant cognitive impairments accompanied by increased expressions in interleukin-1 beta and caspase-3, and decreased expressions in BDNF, phospho-TrkB (pTrkB), postsynaptic density 95, and synapses, which were reversed by VPA. However, TrkB antagonist K252a abolished the beneficial effects of VPA with regard to cognition and decreased pTrkB expression and synapses in the hippocampus. Taken together, the findings of the present study suggested chronic treatment with VPA reverses cognitive deficits through mechanisms probably via a reduction in inflammation and apoptosis in the brain, as well as the activation of the BDNF-TrkB signaling pathway in a mouse model of SAE.

Summary: The study aimed to assess the role of propofol on osteosarcoma and explore the possible mechanisms. The results showed that propofol could inhibit the proliferation and invasion of osteosarcoma and increase the sensitivity of doxorubicin. The experimental results also demonstrated that propofol could induce ER stress and exert an anti-tumor effect.

Key role of the gut-microbiota-brain axis via the subdiaphragmatic vagus nerve in demyelination of the cuprizone-treated mouse brain

Summary: Multiple sclerosis (MS) is a common demyelinating disease that affects the central nervous system. This study investigated the effects of subdiaphragmatic vagotomy (SDV) on demyelination in cuprizone (CPZ)-treated mice and found that SDV significantly improved demyelination and microglial activation in the brain. Analysis of gut microbiota composition and blood metabolites revealed that SDV also improved the abnormal levels in CPZ-treated mice. These findings suggest a link between the gut microbiota-brain axis and demyelination in the brain.

Integrated Proteomic and Phosphoproteomic Analysis of the Hippocampus in a Mouse Model of Early Life Inflammation

Summary: This study investigated the potential molecular mechanisms of early life inflammation-induced cognitive impairment by conducting proteomic and phosphoproteomic analysis of the hippocampus. It was found that early life lipopolysaccharide (LPS) exposure led to cognitive impairment in male mice but not in female mice. The analysis also revealed that early life LPS exposure affected the glutamatergic synapse and neuroactive ligand-receptor interaction, which are associated with synaptic function and energy metabolism.

Effect of Remimazolam on Postoperative Delirium in Older Adult Patients Undergoing Orthopedic Surgery: A Prospective Randomized Controlled Clinical Trial

Summary: This randomized controlled trial aimed to evaluate the relationship between remimazolam and postoperative delirium in older adult patients undergoing orthopedic surgery. The incidence of postoperative delirium in the remimazolam group was 15.6%, while it was 12.4% in the propofol group, with no significant difference observed. Although patients in the remimazolam group had a lower incidence of hypotension and consumed fewer vasoactive drugs intraoperatively, they experienced longer postoperative extubation time and PACU stay.

Predicting postoperative gastric cancer prognosis based on inflammatory factors and machine learning technology

Summary: Machine learning can predict gastric cancer patients' prognosis, but no studies have explored predicting gastric cancer death based on machine learning using related inflammatory factor variables. In this study, six machine learning algorithms were applied to predict total gastric cancer death after surgery, and the neutrophil-lymphocyte ratio, platelet lymphocyte ratio, and age were found to be the most important factors.

Risk of esketamine anesthesia on the emergence delirium in preschool children after minor surgery: a prospective observational clinical study

Summary: This study investigated the effects of a single-dose of esketamine during anesthesia induction on emergence delirium (ED) in preschool children after minor surgery. The study found that the use of esketamine was associated with a higher incidence and score of ED, as well as a longer post-anesthesia care unit stay. However, there were no significant differences in other outcomes between the two groups. Therefore, the use of esketamine in preschool children for minor surgery should be noticed.

Abnormal compositions of gut microbiota and metabolites are associated with susceptibility versus resilience in rats to inescapable electric stress

Summary: This study found that gut microbiota and microbiome-derived metabolites play important roles in susceptibility versus resilience in rats exposed to inescapable electric stress. The relative abundances of Asaccharobacter, Eisenbergiella, and Klebsiella in the gut microbiota of susceptible rats were significantly higher than that of resilient rats. Additionally, several microbiome and metabolites were found to be significantly altered between susceptible and resilient rats.

HMGB1 mediates synaptic loss and cognitive impairment in an animal model of sepsis-associated encephalopathy

Plasticity in ventral pallidal cholinergic neuron-derived circuits contributes to comorbid chronic pain-like and depression-like behaviour in male mice

Summary: The study focuses on the role of ventral pallidum (VP)-derived cholinergic circuits in hyperalgesia and depression-like behavior in a chronic pain mouse model. The cholinergic circuits are activated by pain-like stimuli and become hyperactive during persistent pain. Acute stimulation of these circuits reduces pain threshold, while inhibition of the circuits elevates pain threshold. Modulation of the VP-derived cholinergic pathway for multiple days regulates depression-like behavior in chronic pain. Therefore, these circuits are implicated in comorbid hyperalgesia and depression-like behavior in a chronic pain mouse model.

Melanin-like polydopamine nanoparticles mediating anti-inflammatory and rescuing synaptic loss for inflammatory depression therapy

Summary: Inflammatory depression is closely related to neuroinflammation. Melanin-like polydopamine nanoparticles (PDA NPs) were synthesized using nano-based drug delivery technology to overcome the low permeability of current anti-inflammatory drugs. PDA NPs, with abundant phenolic hydroxyl groups, showed excellent antioxidative properties and were effective in treating lipopolysaccharide-induced inflammatory depression in mice. PDA NPs reduced inflammation, restored synaptic loss, and exhibited good biocompatibility.
