Sulbutiamine Counteracts Trophic Factor Deprivation Induced Apoptotic Cell Death in Transformed Retinal Ganglion Cells

Sulbutiamine is a highly lipid soluble synthetic analogue of vitamin B-1 and is used clinically for the treatment of asthenia. The aim of our study was to demonstrate whether sulbutiamine is able to attenuate trophic factor deprivation induced cell death to transformed retinal ganglion cells (RGC-5). Cells were subjected to serum deprivation for defined periods and sulbutiamine at different concentrations was added to the cultures. Various procedures (e.g. cell viability assays, apoptosis assay, reactive oxygen species analysis, Western blot analysis, flow cytometric analysis, glutathione (GSH) and glutathione-S-transferase (GST) measurement) were used to demonstrate the effect of sulbutiamine. Sulbutiamine dose-dependently attenuated apoptotic cell death induced by serum deprivation and stimulated GSH and GST activity. Moreover, sulbutiamine decreased the expression of cleaved caspase-3 and AIF. This study demonstrates for the first time that sulbutiamine is able to attenuate trophic factor deprivation induced apoptotic cell death in neuronal cells in culture.

Summary: This study evaluated the cytoprotective, anti-mutagenic, and anticlastogenic activities of standardized Orthosiphon stamineus extract. The extract showed potent scavenging effects on hydrogen peroxide and other free radicals, stimulated glutathione production in liver cells, and exhibited powerful antimutagenic activities. Cytogenetic tests showed reduced mitotic and replicative indices without any chromosomal abnormalities or sister chromatid exchange. Pretreatment with the extract proved to be an efficient protocol in ameliorating the effects of mitomycin C.

Summary: Through a combination of spectroscopic and crystallographic methods, natural compounds with different chemical structures were isolated from Juniperus chinensis, with novel compounds being photochemically produced. One of the compounds significantly inhibited the activity of tyrosinase as predicted by a machine learning-based assay, while also displaying characteristics of inhibiting lipid accumulation and enhancing extracellular glycerol excretion.

Natural Photosensitizers in Antimicrobial Photodynamic Therapy

Caenorhabditis elegans as a powerful tool in natural product bioactivity research

Summary: Apart from synthetic and semisynthetic compounds, natural products are important sources of potential therapeutic lead compounds for various diseases. In vivo assays, particularly using Caenorhabditis elegans, provide valuable insights into the efficacy, adverse effects, and modes of action of these candidates. This review highlights commonly used techniques for screening the bioactivities of natural products, and explores recent studies on their applications in anti-aging, anti-neurodegeneration, anti-obesity, anti-infection, and gut health using the C. elegans model. Technological developments in instrumental analysis and emerging methods involving C. elegans are also discussed.

3,3′-Diindolylmethane Supplementation Maintains Oocyte Quality by Reducing Oxidative Stress and CEP-1/p53-Mediated Regulation of Germ Cells in a Reproductively Aged Caenorhabditis elegans Model

Summary: In this study, the effects of 3,3'-diindolylmethane (DIM) on oocyte quality and reproductive aging were investigated in a model organism. DIM was found to improve mitochondrial function, reduce oxidative stress, and decrease chromosomal aberrations in aged oocytes, leading to improved fertility. Additionally, DIM supplementation maintained proper levels of germ cell apoptosis and proliferation, further delaying the onset of reproductive aging.

Antimicrobial photodynamic therapy with Ligularia fischeri against methicillin-resistant Staphylococcus aureus infection in Caenorhabditis elegans model

Summary: This study evaluated the antibacterial properties of antimicrobial photodynamic therapy combined with L. fischeri extract against MRSA and various pathogens, and assessed its photopharmaceutical actions in C. elegans. The results showed that this therapy effectively inhibited the growth of pathogens and improved the quality of life and lifespan in MRSA-infected worms without causing severe side effects. These findings highlight the significance of this treatment for safe MRSA phototreatment in biomedical and cosmeceutical industries.
