Summary: This study explored the effect of phlorizin on cholinergic memory impairment and dysbacteriosis in D-galactose induced ICR mice. The results showed that phlorizin could reverse the loss of organ weight and body weight caused by D-galactose, and had effects on improving cognitive function and anti-inflammatory and antioxidant effects. In addition, phlorizin could improve the state of the hippocampus and cecum by regulating gene expression, increasing acetylcholine levels, inhibiting acetylcholinesterase activity, and rebuilding gut microbiota balance by increasing short-chain fatty acids and reducing lipopolysaccharide levels.

Summary: Galantamine, one of the approved agents for Alzheimer's disease, acts as an inhibitor of acetylcholinesterase and a possible positive allosteric modulator of neuronal alpha 7 nicotinic acetylcholine receptors. The deconstructed analogs showed that the cyclohexa(e)nyl ring and/or hydroxyl group are important for the agonist action inhibition and optimal activity. However, the deconstructed analogs were less potent than galantamine as AChE inhibitors.

Summary: Small doses of oral D-glyceric acid (DGA) have been shown to activate mitochondrial metabolism and reduce inflammation. The present study reveals that after a 4-day DGA regimen, a dose of DGA can acutely activate the HO-1 pathway, while long-term DGA regimen may downregulate the HO-1 pathway. These findings are important for the prevention of chronic diseases, especially aging-related diseases.

In this study, in silico and machine learning techniques were used to predict binding energy and evaluate analogues of ASEM.

Summary: This study examined the effects of TaPcZn-PDT on cytotoxicity, cell viability, apoptosis, pyroptosis, cellular reactive oxygen species (ROS), mitochondrial membrane potential (??m), caspase-1, caspase-3, and nuclear transcription factor-?B (NF?B) in MCF-7 cells. The results showed that TaPcZn-PDT increased cytotoxicity and the percentage of apoptotic and pyroptotic cells, decreased cell viability and ??m, induced ROS production, and activated caspase-1, caspase-3, and NF?B in MCF-7 cells. siRNA targeting caspase-1 attenuated the effects of TaPcZn-PDT on apoptosis, pyroptosis, and the activation of caspase-1, caspase-3, and NF?B in MCF-7 cells. Therefore, caspase-1 plays a regulatory role in the apoptosis and pyroptosis induced by TaPcZn-PDT in MCF-7 cells.
