ALZHEIMER DISEASE New light on an old CLU

Genome-wide association studies (gwas) have uncovered over two dozen candidate Alzheimer disease susceptibility genes; however, the results of these studies showed limited overlap. Two independently performed gwas involving cohorts from Europe and the us have now identified three additional putative Alzheimer disease genes that show modest but remarkably consistent effects across data sets.

Summary: The levels of total tau (t-tau) in cerebrospinal fluid (CSF) in AD are associated with changes in proteins related to neuronal plasticity and dysfunction of the blood-brain and blood-CSF barriers. These protein changes are disease traits and may affect treatment response. Therefore, future research may need to stratify based on CSF t-tau levels.

Summary: We found heterogeneity in regional-atrophy patterns through latent-profile analysis of 1,482 longitudinal magnetic resonance imaging observations. The results supported a 2-group solution reflecting differences in atrophy rates in cortical regions and hippocampus along with comparable caudate atrophy. Our findings suggest distinct mechanisms of atrophy in striatal versus hippocampal-cortical systems.

A comparison between similarity matrices for principal component analysis to assess population stratification in sequenced genetic data sets

Summary: Genetic similarity matrices are commonly used to assess population substructure, and in this study, the performance of three different matrices was evaluated. The results showed that rare single-nucleotide variations (SNVs) provided the best clustering performance, with the unweighted Jaccard matrix being the most preferable for rare variants. The visual inspection and clustering metrics confirmed its superiority compared to other methods and different allele frequency cutoffs. The application of these matrices to Alzheimer's disease data sets and different populations revealed the role of rare variants in local and global population substructure.

Summary: Dobricic et al. quantified the expression of 10 candidate microRNAs for Parkinson's disease and Alzheimer's disease and found that hsa-miR-132-3p, hsa-miR-132-5p, and hsa-miR-129-5p were differentially expressed in both diseases, suggesting shared pathogenic mechanisms. This study provides valuable insights into the dysregulation of microRNA expression in neurodegenerative diseases, and highlights the potential of these microRNAs as therapeutic targets.
