A CRISPR/Cas9-Engineered ARID1A-Deficient Human Gastric Cancer Organoid Model Reveals Essential and Nonessential Modes of Oncogenic Transformation

Yuan-Hung Lo, Kevin S. Kolahi, Yuhong Du, Chiung-Ying Chang, Andrey Krokhotin, Ajay Nair, Walter D. Sobba, Kasper Karlsson, Sunny J. Jones, Teri A. Longacre, Amanda T. Mah, Bahar Tercan, Alexandra Sockell, Hang Xu, Jose A. Seoane, Jin Chen, Ilya Shmulevich, Jonathan S. Weissman, Christina Curtis, Andrea Califano, Haian Fu, Gerald R. Crabtree, Calvin J. Kuo

Summary: Mutations in ARID1A are common in human cancer, but the oncogenic consequences of these mutations remain unclear. Using CRISPR/Cas9, researchers induced dysplasia, tumorigenicity, and mucinous differentiation in human gastric organoids, providing insights into the pathways affected by ARID1A mutation. Further analysis showed vulnerability of ARID1A-deficient organoids to inhibition of BIRC5/survivin, highlighting this pathway as crucial for early-stage gastric tumorigenesis.

BAF subunit switching regulates chromatin accessibility to control cell cycle exit in the developing mammalian cortex

Simon M. G. Braun, Ralitsa Petrova, Jiong Tang, Andrey Krokhotin, Erik L. Miller, Yitai Tang, Georgia Panagiotakos, Gerald R. Crabtree

Summary: Loss of the neural development-related BAF subunit BAF53a leads to cell cycle arrest and disruption of neurogenesis due to altered chromatin accessibility, which inhibits the binding of specific neural transcription factors and suppresses cell cycle gene expression, highlighting a novel mechanism regulating neural progenitor cell cycle exit in the presence of extrinsic proliferative cues.

Recent findings suggest adding red fox (Vulpes vulpes) to climate- threatened whitebark pine (Pinus albicaulis) trophic system

Patrick R. Cross, Robert L. Crabtree

Summary: Mountain ecosystems are sensitive to climate change, and the decline in whitebark pine seed production can impact food webs. This study found that whitebark pine seed availability affects winter foraging and habitat use by Rocky Mountain red foxes, with consumption and use decreasing during non-mast winters. Further research is needed to understand the importance of whitebark pine seeds for montane red foxes and to consider it in conservation efforts.

Electronic and Spin-State Effects on Dinitrogen Splitting to Nitrides in a Rhenium Pincer System

Jeremy E. Weber, Faraj Hasanayn, Majed Fataftah, Brandon Q. Mercado, Robert H. Crabtree, Patrick L. Holland

Summary: The study reports a new type of N-2-bridged rhenium (III) complex in which N-2 binding occurs at a higher oxidation level. Computational studies show that the weakly pi-donor pincer ligand stabilizes an open-shell electronic configuration, leading to enhanced N-2 binding in the bridged complex. The SQUID magnetometry analysis confirms a singlet ground state for this complex and offers explanations for the antiferromagnetic coupling observed.

Distorted Copper(II) Complex with Unusually Short CF•••Cu Distances

Claire C. Cody, H. Ray Kelly, Brandon Q. Mercado, Victor S. Batista, Robert H. Crabtree, Gary W. Brudvig

Summary: The Cu(II)-(L-CF3)(2) complex exhibits a distorted seesaw geometry with the shortest crystallographic CF center dot center dot center dot Cu distances reported to date, likely due to specific ligand interactions. This distortion may have a steric origin according to comparisons with other versions of the complex.

Accessing Molecular Dimeric Ir Water Oxidation Catalysts from Coordination Precursors

Gongfang Hu, Jennifer L. Troiano, Uriel T. Tayvah, Liam S. Sharninghausen, Shashi Bhushan Sinha, Dimitar Y. Shopov, Brandon Q. Mercado, Robert H. Crabtree, Gary W. Brudvig

Summary: One ongoing challenge in the field of iridium-based water oxidation catalysts is to develop a molecular precatalyst affording well-defined homogeneous active species for catalysis. A new dinuclear iridium catalyst has been described in this study, which exhibits comparable oxygen-evolution activity without the need for oxidative activation.

mSWI/SNF promotes Polycomb repression both directly and through genome-wide redistribution

Christopher M. Weber, Antonina Hafner, Jacob G. Kirkland, Simon M. G. Braun, Benjamin Z. Stanton, Alistair N. Boettiger, Gerald R. Crabtree

Summary: Research shows that rapid depletion of BAF redistributes Polycomb repressive complexes, leading to decompaction of highly repressed domains and transcriptional derepression. By degrading PRC1 and PRC2, a traditional role for BAF in Polycomb-mediated repression was identified, along with global Polycomb redistribution.

Systemic enhancement of serotonin signaling reverses social deficits in multiple mouse models for ASD

Jessica J. Walsh, Pierre Llorach, Daniel F. Cardozo Pinto, Wendy Wenderski, Daniel J. Christoffel, Juliana S. Salgado, Boris D. Heifets, Gerald R. Crabtree, Robert C. Malenka

Summary: Autism spectrum disorder (ASD) is a heterogeneous neurodevelopmental disorder caused by various genetic and environmental factors, characterized by social interaction impairments. Research has shown that enhancing 5-HT activity can reverse social deficits in ASD mouse models, while MDMA and a 5-HT1b receptor agonist are effective in rescuing sociability deficits in multiple ASD mouse models.

Optimization of Surface Loading of the Silatrane Anchoring Group on TiO2

Jennifer L. Troiano, Robert H. Crabtree, Gary W. Brudvig

Summary: Anchoring groups are important for attaching small molecules to metal oxide surfaces, such as in water-splitting dye-sensitized photoelectrochemical cells (WS-DSPECs). In this study, the surface loading of the silatrane anchoring group on titanium dioxide surfaces was optimized. The researchers found that previous methods resulted in low surface coverage, but they were able to uncover factors contributing to this issue and develop methods to greatly improve surface coverage, particularly through the use of a benzoic acid additive.

Organometallic complexes as preferred precursors to form molecular Ir(pyalk) coordination complexes for catalysis of oxygen evolution

Gongfang Hu, Robert H. Crabtree, Gary W. Brudvig

Summary: The study found that organometallic complexes can be effective catalyst precursors even if the organometallic ligands are lost during the activation process, potentially suppressing pathways that lead to undesirable nanoparticle production.

Increased ACTL6A occupancy within mSWI/SNF chromatin remodelers drives human squamous cell carcinoma

Chiung-Ying Chang, Zohar Shipony, Sherry G. Lin, Ann Kuo, Xiaochen Xiong, Kyle M. Loh, William J. Greenleaf, Gerald R. Crabtree

Summary: Overexpression of ACTL6A leads to its stoichiometric assembly into BAF complexes and enhances interaction with specific regulatory regions in the genome, activating SCC genes. This highlights a molecular AND gate mechanism involving ACTL6A amplification that is critical for SCC initiation and maintenance.
