Summary: Researchers have developed a new approach using metabolic glycan labeling to improve the therapeutic efficacy of dendritic cell vaccines. This approach enhances the antigen presentation and T cell activation of dendritic cells, as well as the cytotoxic T lymphocyte response and antitumor efficacy. The labeling method also enables the conjugation of cytokines onto adoptively transferred dendritic cells to further enhance their immune response.

Summary: Therapeutic cancer vaccines have shown clinical benefit in mCRPC patients, but the effects of sequential checkpoint inhibitors after therapeutic vaccine are not well understood. A study using avelumab as anti-PD-L1 therapy in mCRPC patients with previous therapeutic vaccine showed disappointing results, indicating a poor response in this patient population.

Summary: This study identified cardiovascular adverse effects associated with sipuleucel-T, including myocardial ischemia, supraventricular tachycardia, congestive heart failure, and valvular disorders. Patients with cardiovascular risk factors receiving sipuleucel-T may be at higher risk for congestive heart failure, myocardial ischemia, and supraventricular tachycardia, suggesting potential T-cell-mediated cardiovascular toxicity with cancer immunotherapy.

Dendritic cell vaccine therapy for colorectal cancer

Summary: Current standard-of-care interventions for colorectal cancer include surgical resection, chemotherapy, and targeted therapies, with the combination of anti-vascular endothelial growth factor agents and chemotherapy being an important development. Dendritic cell vaccines targeting tumor-derived blood vessels may disrupt tumor angiogenesis and decrease tumor growth, but clinical results have historically been disappointing. The success of Sipuleucel-T in prostate cancer patients has revived the clinical development of dendritic cell vaccines for other types of cancer, including colorectal cancer.

Through analysis of gene expression in prostate epithelial cells, four distinct subtypes of prostate adenocarcinoma were identified: luminal A, luminal S, AVPC-I, and AVPC-M, each with unique characteristics and drug resistance patterns. The PSA/PAP ratio in advanced cancer may help determine which patients would benefit from specific treatments.

Combination therapy for mCRPC with immune checkpoint inhibitors, ADT and vaccine: A mathematical model

Summary: Metastatic castration resistant prostate cancer (mCRPC) is commonly treated with androgen deprivation therapy (ADT) combined with chemotherapy. Immune therapy using checkpoint inhibitors has shown promising results in other cancers but initial clinical trials in mCRPC have been disappointing. This paper presents a mathematical model that simulates the effects of different treatment combinations and doses, demonstrating that larger doses of checkpoint inhibitors can reduce tumor volume. Therefore, identifying optimal treatment combinations and doses for individual patients is crucial.
