Summary: Genetic and epigenetic events contribute to the downregulation of the cellular antigen processing and presentation machinery (APM), leading to cancer evasion of the immune system. A high-throughput cell-based screening assay has identified curcuphenol, a compound derived from turmeric, as a potential reversal agent for the downregulation of APM components in metastatic tumour cells. The findings suggest that curcuphenol can induce the expression of APM components and potentially be used in the development of therapeutic candidates for metastatic cancers.

Further analysis revealed a reverse-binding mode in HLA-DP ligands, and a machine-learning framework was developed to accurately predict binding specificities and ligands of any MHC-II allele.

Summary: Major Histocompatibility Complex (MHC) Class II deficiency is a rare autosomal recessive primary immunodeficiency caused by mutations in regulatory genes of MHC Class II proteins. This case report describes a 9-month-old girl presenting with repeated pneumonia, lymphocytopenia, and absence of HLA-DR expression on B cells, which led to the diagnosis of MHC Class II deficiency. Genetic analysis identified a homozygous mutation in the RFX-5 gene. Despite the patient's unfortunate outcome, the importance of prenatal diagnosis was emphasized to the parents.

Summary: This study investigated the mechanism behind the reduced allergenicity of shrimp (Macrobrachium nipponense) when subjected to combined thermal/pressure processing. The findings revealed that mice treated with steamed + reverse-pressure sterilized shrimp exhibited lower allergic reactions compared to those fed with raw shrimp or steamed shrimp. This reduction in allergenicity was attributed to protein unfolding, exposure of hydrophobic residues, disruption of immunodominant linear epitopes, and the heat/digestion stability of certain epitopes.

Summary: The study demonstrates that IAV infection induces endogenous presentation of a viral epitope by MHC-II to CD4(+) T cells, dependent on de novo protein synthesis and the ER-Golgi network. Some antigenic peptides are of cytosolic origin in this process. Additionally, autophagy inhibitors and deletion of autophagy-related genes block most of the MHC-II-restricted endogenous IAV antigen presentation.

Summary: The therapy of cancer by adoptive T cell transfer (ACT) requires T cell receptors (TCRs) with optimal affinity for HLA class I-bound peptides (pHLA-I). But not every patient responds to ACT. This study investigated the contribution of ERAP1 and ERAP2 to TCR recognition of cancer cells and found that ERAP2 plays a significant role in tumor epitope presentation and TCR recognition. These findings may have implications for future ACT design.

Summary: The liver is a tolerogenic organ that can induce immune tolerance against self-antigens. However, autoimmune liver diseases can disrupt this tolerance due to genetic predisposition, environmental factors, and imbalance in regulatory mechanisms. Liver cells, including conventional and unconventional antigen presenting cells (APCs), present self-antigens to T cells, leading to altered immune responses, liver injury, and inflammation. Understanding the impact of antigen presentation and the role of immune cells in liver inflammation could aid in the development of novel therapeutic strategies for autoimmune liver diseases.
