Chemokine Receptors CC Chemokine Receptor 5 and C-X-C Motif Chemokine Receptor 4 Are New Therapeutic Targets for Brain Recovery after Traumatic Brain Injury

Sigal Liraz-Zaltsman, Yael Friedman-Levi, Dalia Shabashov-Stone, Galit Gincberg, Dana Atrakcy-Baranes, Mary Teena Joy, S. Thomas Carmichael, Alcino J. Silva, Esther Shohami

Summary: Inhibition of CCR5 signaling has been shown to have neuroprotective effects after traumatic brain injury, reducing lesion size, inflammation, and increasing ERK phosphorylation. Increased expression of both CCR5 and CXCR4 was observed in cortical cells 3 to 11 days post-injury, suggesting a potential therapeutic window for intervention.

Pharmacological blockers of CCR5 and CXCR4 improve recovery after traumatic brain injury

Yael Friedman-Levi, Sigal Liraz-Zaltsman, Chen Shemesh, Kinneret Rosenblatt, Efrat L. Kesner, Galit Gincberg, S. Thomas Carmichael, Alcino J. Silva, Esther Shohami

Summary: Blocking CCR5 and CXCR4 after closed head injury (CHI) enhances tissue preservation, increases levels of synaptic cognition-related signaling molecules, and promotes neurobehavioral recovery. These findings suggest that the drugs used in this study, which are FDA approved and in clinical use, should be considered for translational research in TBI patients.

Dorsal premammillary projection to periaqueductal gray controls escape vigor from innate and conditioned threats

Summary: The study showed that PMd-cck cells in mice are activated during escape, and inhibition of the PMd decreases escape speed. The PMd-dlPAG circuit is identified as a central node controlling escape behavior in response to threats.

Postnatal immune activation causes social deficits in a mouse model of tuberous sclerosis: Role of microglia and clinical implications

Manuel F. Lopez-Aranda, Ishanu Chattopadhyay, Gayle M. Boxx, Elizabeth R. Fraley, Tawnie K. Silva, Miou Zhou, Miranda Phan, Isaiah Herrera, Sunrae Taloma, Rochelle Mandanas, Karen Bach, Michael Gandal, Daniel H. Geschwind, Genhong Cheng, Andrey Rzhetsky, Stephanie A. White, Alcino J. Silva

Summary: Research indicates that early postnatal immune activation has a significant impact on social memory and behavior, particularly in male mice heterozygous for the gene responsible for tuberous sclerosis complex. These effects are caused by abnormal interferon signaling and microglia function, and are associated with an increased risk of autism spectrum disorders in males with a history of hospitalizations due to infections.

Miniscope-LFOV: A large-field-of-view, single-cell-resolution, miniature microscope for wired and wire-free imaging of neural dynamics in freely behaving animals

Changliang Guo, Garrett J. Blair, Megha Sehgal, Federico N. Sangiuliano Jimka, Arash Bellafard, Alcino J. Silva, Peyman Golshani, Michele A. Basso, Hugh Tad Blair, Daniel Aharoni

Summary: We have developed an open-source miniature microscope called MiniLFOV for neural imaging in freely behaving rats. It offers a large field of view and cellular resolution imaging, with the capability of deep brain and cortical imaging. The MiniLFOV has been successfully validated in both rats and head-fixed mice, and it also supports wire-free operation with a novel data acquisition expansion board. We believe that this new implementation of the UCLA Miniscope platform will greatly benefit researchers in studying brain function in freely behaving animals.

Experiment Selection in Meta-Analytic Piecemeal Causal Discovery

Nicholas J. Matiasz, Justin Wood, Wei Wang, Alcino J. Silva, William Hsu

Summary: Scientists aim to design experiments for maximal information, sometimes needing to choose between intervening on variables and observing responses. Previous work developed software for annotating literature statistics and generating causal explanations, aiding evidence synthesis and experiment planning. Introduces interpretable experiment selection policies in piecemeal causal discovery context, showing efficiency in identifying causal structures through simulations.
