Summary: The Cu-64-DOTA-ECL1i PET uptake correlated with CCR2(+) cell infiltration in the lung associated with fibrosis, with inhibition of fibrosis by IL1 beta blockade or pirfenidone reducing CCR2(+) macrophage accumulation. CCR2(+) cells concentrated in perifibrotic regions in lung tissues from patients with IPF, exhibiting intensive signals in fibrotic zones compared to healthy volunteers. These findings suggest a role for imaging CCR2(+) cells within the fibrogenic niche in IPF for personalized therapy and monitoring.

The immune landscape in tuberculosis reveals populations linked to disease and latency

Ekaterina Esaulova, Shibali Das, Dhiraj Kumar Singh, Jose Alberto Choreno-Parra, Amanda Swain, Laura Arthur, Javier Rangel-Moreno, Mushtaq Ahmed, Bindu Singh, Ananya Gupta, Luis Alejandro Fernandez-Lopez, Maria de la Luz Garcia-Hernandez, Allison Bucsan, Chivonne Moodley, Smriti Mehra, Ethel Garcia-Latorre, Joaquin Zuniga, Jeffrey Atkinson, Deepak Kaushal, Maxim N. Artyomov, Shabaana A. Khader

Summary: The immune landscape of the lungs in NHPs with LTBI and PTB was characterized using high-throughput technologies. PTB in macaque lungs is characterized by the influx of pDCs, an IFN-responsive macrophage population, and activated T cell responses. On the other hand, LTBI in macaques is marked by the accumulation of a CD27(+) NK cell subset in the lungs.

Summary: This study suggests that the age dependency of childhood asthma is related to the evolving host response to respiratory viral infection. The research shows that the traditional age gradient in pediatric asthma exacerbations was disrupted during the coronavirus disease 2019 lockdown, indicating a connection between asthma remission and virus exposure. The study also highlights the role of age-specific responses, particularly in the type 2 inflammatory reactions to viruses.

Identification of kidney injury-released circulating osteopontin as causal agent of respiratory failure

Fatima Zohra Khamissi, Liang Ning, Eirini Kefaloyianni, Hao Dun, Akshayakeerthi Arthanarisami, Amy Keller, Jeffrey J. Atkinson, Wenjun Li, Brian Wong, Sabine Dietmann, Kory Lavine, Daniel Kreisel, Andreas Herrlich

Summary: This study identifies circulating osteopontin (OPN) released from kidney tubule cells as a novel mediator in acute kidney injury (AKI)-induced acute lung injury (ALI). OPN triggers lung endothelial leakage, inflammation, and respiratory failure. The study also demonstrates the elevated levels of OPN in AKI patients and its correlation with kidney injury.

Macrophage NOX2 NADPH oxidase maintains alveolar homeostasis in mice

Sourav Bhattacharya, Rachel A. Idol, Wei Yang, Jorge David Rojas Marquez, Yanan Li, Guangming Huang, Wandy L. Beatty, Jeffrey J. Atkinson, John H. Brumell, Juhi Bagaitkar, Jeffrey A. Magee, Mary C. Dinauer

Summary: NOX2 plays a role in maintaining lung homeostasis and suppressing alveolar macrophage activation. The absence of NOX2 leads to enhanced cytokine responses in macrophages and lung inflammation.
